摘  要:目的:通过摇瓶发酵筛选出土曲霉(A.terreus)ATCC 20542生物合成洛伐他汀所需的最适碳源和氮源。方法:保持发酵培养基中其他组分不变,分别以麦芽糊精、葡萄糖、乳糖、淀粉作为碳源,以蛋白胨、酵母粉、硫酸铵、大豆粉为氮源进行发酵,并以乙酸乙酯萃取发酵液中的洛伐他汀,用高效液相色谱法(HPLC)对其进行相对定量。结果:四种碳源中,淀粉产生的洛伐他汀的量最大;四种氮源中,大豆粉产生的洛伐他汀的量最大。结论:土曲霉生产洛伐他汀的培养基中最佳碳源为淀粉,最佳氮源为大豆粉。Objective : To screen the optimal carbon and nitrogen sources of fermentation medium for lovastatin produced by Aspergillus terreus ATCC 20542. Method: With other components of the medium unchanged, maltodextrin, glucose, lactose, starch were tested as the carbon sources, while peptone, yeast powder, ammonium sulfate, soybean powder were tested as the ni- trogen sources. Lovastatin was extracted from the broth by acetoacetate and was measured by high performance liquid chromatog- raphy (HPLC). Result: More lovastatin was produced when starch and soybean powder were used as the carbon and nitrogen sources, respectively. Conclusion: In fermentation medium for lovastatin produced by Aspergillus terreus, starch was the best carbon source, and soybean powder was the best nitrogen source.
