摘 要:目的探讨纳米级磁性阿霉素脂质体的制备及其体外细胞毒性。方法用反相蒸发法制备纳米级磁性阿霉素脂质体,以正交实验设计确定最佳制备工艺;电镜观察其形态,Zetapal无机粒度分析仪测量其粒径分布,利用紫外-Sephadex G-50法测定其包封率,利用磁测仪测量其磁响应性。用比色法分析其对人胃癌细胞的体外细胞毒作用。结果磁性阿霉素脂质体在电镜下为圆形或椭圆形,平均粒径为(293.4±20.6)nm,包封率为(30.8±3.3)%,磁响应性50.28emu,其体外抑制肿瘤细胞生长的能力与阿霉素无显著差异。结论用反相蒸发法制备的磁性阿霉素脂质体平均粒径小,达到纳米级,磁响应性高,抑瘤效果好。Objective To explore the preparation of magnetic doxorubicin(DOX) nanoliposomes and their cytotoxicity in vitro.Methods Magnetic DOX liposomes were prepared by the reverse-phase evaporation method,an optimal technique which was chosen with orthogonal design,and their diameter distribution was observed under an electronic microscope.The encapsulation efficiency was detected by using sephadex column-ultraviolet method.The magnetic properties of the magnetic DOX liposomes were measured by using a magnetometer.The MTT assay was used to measure the chemosensitivity of human gastric neoplasm cells to magnetic DOX liposomes.Results The magnetic DOX liposomes were globular or elliptic,the average particle size was(293.4±20.6) nm,their encapsulation efficiency was(30.8±3.3)%,and their average magnetic response was 50.28 emu.There was no significant difference in restraining growth of gastric neoplasm cells between the magnetic DOX liposomes and DOX.Conclusion The magnetic DOX liposomes had less diameter and higher magnetic response that could get approving effect to restrain the growth of neoplasm cells.
