摘  要:目的研究天然化合物青藤碱对人前列腺癌DU145细胞增殖及凋亡的影响。方法采用MTT法检测青藤碱对DU145细胞增殖的影响;采用流式细胞仪AnnexinV/PI双染法检测青藤碱对DU145细胞凋亡的影响;采用电子显微镜观察青藤碱作用DU145细胞后细胞超微结构的变化。结果 MTT结果显示青藤碱浓度在100~400μmol/L时,作用24、48小时后,细胞增殖受到抑制,呈现明显时间剂量效应关系;流式细胞仪AnnexinV/PI双染法显示青藤碱(0、200、400μmol/L)作用于细胞24小时后,细胞凋亡率升高,且呈剂量效应关系;电子显微镜下青藤碱处理组细胞可见染色质浓聚、边集等典型的凋亡改变,而空白对照组没有相应的变化。结论青藤碱能有效抑制人前列腺癌DU145细胞增殖、诱导细胞凋亡。Objective To investigate the effects of sinomenine on cell proliferation and apoptosis in DU145 prostate cancer cells. Method Cell proliferation was measured using the MTT assay. Induction of apoptosis was determined by flow cytometry analysis using an Annexin V-FITC Apoptosis Kit. The uhrastructure of sinomenine-treated DU145 cells was investigated by transmission electron microscopy. Result The cells were treated with sinomenine (100 -400 μmol/L) for 24 and 48 h,the growth of DU145 cells was suppressed in a dose-and time-dependent manner(P〈 0.05). Flow cytometric analysis showed that sinomenine induced DU145 cells apoptosis in a dose-dependent manner(P〈0.05 ). Chromosome condensation was observed by transmission electron microscopy in treated DU145 cells, while the untreated cells exhibited a fairly intact morpho!ogy. Conclusion Sinomenine could inhibit the growth of DU145 prostate cancer cells and induce cell apoptosis.
