Wnt/β-catenin信号通路激活诱导食管癌Eca-109细胞miRNA表达谱的研究

摘　　要：目的:探讨Wnt/β-catenin信号通路对食管鳞癌Eca-109细胞系miRNA表达的影响,为靶向该通路相关miRNA治疗提供实验依据。方法:食管癌Eca-109细胞分别以培养基终浓度10ng/ml、20ng/mlWNT3a处理48小时,细胞免疫荧光、Western-blot、RT-PCR检测β-catenin表达水平;芯片检测WNT3a处理组和对照组miRNA表达谱;qRT-PCR验证miRNAs表达水平。结果:WNT3a可上调食管癌细胞β-cate-nin mRNA和蛋白水平,增加其细胞核内分布;芯片检测经qRT-PCR验证,WNT3a处理后食管癌Eca-109细胞相对于未处理组miR表达改变,其中miR-21(3.87倍,P=0.002)、miR-638(2.90倍,P=0.016)显著升高;miR-107(0.18倍,P=0.003)、miR-99b(0.33倍,P=0.019)明显降低,差异均具有统计学意义。结论:Wnt/β-catenin信号通路激活影响食管癌Eca-109细胞miRNAs表达,提示Wnt/β-catenin信号通路的功能也可能通过调控相关miRNA表达参与实现。Objective：To investigate WNT3a induced WNT signal activation affected on miRNA expression in esophageal cancer cell Eca- 109. Methods ：Eca - 109 cells, respectively, cultured in 10ng/ml, 20ng/ml final concen- tration WNT3a in culture medium for 48 hours. By using immunofluorence ,Western -blot and RT- PCR to detect β - catenin expression level to identify WNT pathway activity status, miRNA chip detecting miRNA expression profiling in the control group and 101xg/ml WNT3a- treated cells, and qRT- PCR validation. Results： WNT3a could upregu- late the β - catenin mRNA and protein expression, also increased the nuclear distribution of β - eatenin protein, miR- NA chip detecting and validated by qRT -PCR, WNT3a treated Eca- 109 cell comparing to control, miR -21 （3.87 fold,P =0. 002） ,miR -638（2.90 fold,P =0.016） were significantly increased, miR - 107（0.18 fold,P =0. 003 ）, miR - 99b（0.33 fold, P = 0. 019 ） were significantly decreased, all the differences were statistically significant （ P 〈 0.05 ）. Conclusion： WNT3a induced WNT signal pathway actication could affect on miRNA expression in esophageal cancer Eca- 109 cell line, miRNAs may be involved in implementing the function of Wnt/β -catenin signaling path- ways.
