摘 要:目的:研究通路阻断剂Wortmannin和LiCl对急性髓细胞白血病细胞系Hedgehog/gli通路的调节作用。方法:选取HL-60和K562细胞系,采用Western blot方法检测gli1和gli2蛋白表达,定量PCR检测gli1和gli2 mRNA的变化,WST-1检测不同通路阻断剂联合对细胞增殖的抑制作用。结果:Wortmannin和LiCl均可以上调两细胞系中gli蛋白和mRNA的表达,并可减弱gli阻断剂GANT61对AML细胞系的毒性作用。结论:Wortmannin和LiCl可以上调AML细胞系中Hedgehog通路的表达。Objective:To investigate the regulation of Wortmannin and LiC1 on Hedgehog - gli pathway in AML cell lines. Methods:HL -60 and K562 cell lines were exposed to Wortmannin and LiC1 for 0,12,24 and 48 hours, and the protein and mRNA expression of gli were measured. The apoptosis induced by GANT61 combined with Wort- mannin or GANT61 combined with LiC1 were also measured by WST - 1. Results: Both Wortmannin and LiC1 in- creased the glil and gli2 mRNA expression in H1 -60 and K562 cell lines. And they also increased glil protein ex- pression in HL -60 cell line and gli2 protein expression in K562 cell line. Wortmannin and LiC1 did not induce apop- tosis of HL -60 line. On the contrary they decreased the cytotoxic effect of GANT61 on AML cells. Conclusion:Wort- mannin and LiC1 increase the Hedgehog - gli pathway in AML cell lines and decrease the cytotoxic effect of GANT61.
