大鼠酒精性肝损伤中肝窦毛细血管化发生的研究

摘　　要：目的研究酒精性肝病大鼠模型中肝窦毛细血管化的发生。方法橄榄油拌平衡饲料喂养大鼠的基础上,44只大鼠随机分为2组,染毒组(n=38)给予每日2次白酒、吡唑混合液胃内灌注,分别于第4、8、12和16周末随机处死8只,正常对照组(n=6)给予等量生理盐水每日2次灌胃,于16周全部处死。取肝组织标本,通过苏木素-伊红(HE)染色观察肝脏病理学改变,通过网状纤维染色、Ⅳ型胶原(CⅣ)及层粘连蛋白(LN)免疫组化染色及电子显微镜检查观察肝窦毛细血管化的发生过程。结果随造模时间延长,染毒组依次出现肝组织脂肪变性、气球样变、炎症及纤维化等病理现象,网状纤维沉积逐渐增多(P<0.05),电子显微镜下发现肝窦内皮细胞失窗孔及连续性基底膜形成的现象,肝窦周CⅣ及LN表达逐渐增加(P<0.01)。结论橄榄油拌平衡饲料喂养及白酒、吡唑混合液灌胃可以成功建立酒精性肝病大鼠模型,于建模12周时观察到肝窦毛细血管化形成。Objective To observe the development of hepatic sinusoidal capillarization in alcoholic liver disease （ALD） rats. Methods Forty four rats provided a balanced diet with olive oil were divided into two groups. The rats in exposure group （ n = 38） were orally administered with alcohol and pyrazole, then sacrificed at the end of4th, 8th, 12th and 16th week respectively （n =8）. The rats in control group （n = 6） were given equal physiological saline. Pathological changes in the livers were observed by HE staining. Development of hepatic sinusoidal capillarization was observed with the methods of reticular fiber strains, immunohistochemistry stains of collagen type IV （ C1V ） and laminin （LN）, and electron microscope examination. Results Fatty degeneration, ballooning degeneration, inflammation and fibrosis could be viewed as the modeling extended. Deposition of reticular fibers increased gradually （P 〈 0.05）. Loss of fenestrae in the sinusoidal endothelial cells and the basal membrane forming could be observed under electron microscope. Expressions of C IV and LN surrounding sinusoids increased gradually （P 〈 0.01 ）. Conclusion ALD rat model was established successfully by olive oil diet plus the intragastric administration of alcohol and pyrazol mixture. The formation of hepatic sinusoidal eapillarization was observed at the end of 12th week.
