摘 要:以纤维素膜为基质 ,活化后 ,键合壳聚糖配基 ,制备成亲和层析膜介质 .考察了所制备的膜介质在不同离子强度I、pH、流速条件下对内毒素的去除效果 .结果表明 ,在离子强度为0 1~ 0 4 ,pH值 6~ 8,流速 2~ 6mL/min范围内去除效果最佳 .将此亲和层析膜介质用于去除人血白蛋白溶液、氢化可的松注射液、右旋糖苷 4 0葡萄糖注射液中的内毒素 ,去除率在95%左右 ,用于去除溶菌酶溶液、盐酸丁卡因注射液中的内毒素 ,去除率在 80 %以上 .A cellulose affinity membrane modified by chitosan was prepared The cellulose was activated by potassium periodate, and chitosan was then immobilized on the activated membrane Endotoxin was determinated by a Kinetic-Turbidimetric method using a BET-16 instrument The influence of different ionic strengths, pH values and flow rates on endotoxin removal have been investigated Optimal affinity conditions for endotoxin removal are as follows:ionic strength 0 1~0 4 mol/L, pH 6~8, flow rate 2~6 mL/min Endotoxin can be successfully removed from human serum albumin, hydrocortisone and dextran 40 glucose solution. A removal rate of up to about 95% can be attained Endotoxins can also be removed from lysozyme and tetracaine with the removal rates of over 80% under selected conditions
