摘  要:目的观察姜黄素体外诱导人胃癌SGC7901细胞自噬性凋亡的作用,并探讨其可能的作用机制。方法用不同浓度的姜黄素处理SGC7901细胞,MTT法检测细胞增殖;单丹磺酰尸胺(monodansylcadaverin,MDC)荧光染色和Hoechst33342荧光染色观察细胞自噬和凋亡的发生;流式细胞术检测细胞凋亡率;Westernblot检测细胞凋亡相关蛋白NF-κB、自噬相关蛋白Beclin1及LC3Ⅱ的表达。结果姜黄素可呈剂量依赖性明显抑制SGC7901的生长。MDC染色显示,40μmol/L姜黄素在作用12 h后即可诱导SGC7901细胞发生自噬,24 h自噬减少;Hoechst33342荧光染色显示,40μmol/L姜黄素诱导凋亡在24 h最明显;流式细胞术显示,40μmol/L姜黄素作用24 h时SGC7901细胞凋亡率明显增加。Western blot检测显示,姜黄素诱导SGC7901细胞自噬性凋亡过程中,凋亡相关蛋白NF-κB、自噬相关蛋白Beclin1及LC3的表达均上调。结论姜黄素通过诱导自噬性凋亡抑制人胃癌细胞SGC7901的生长,其机制与NF-κB、Beclin1及LC3蛋白的表达上调有关。【Objective】 To investigate the molecular mechanisms of autophagic apoptosis induced by curcumin in human gastric cancer cell SGC7901 in vivo.【Methods】 The human gastric carcinoma SGC7901 cells were cultured in vivo containing different concentrations of curcumin.Cell viability was analyzed by MTT assay.MDC staining and Hoechst 33342 were used to examine autophagy and apoptosis induced by curcumin.The apoptosis rates were analyzed by flow cytometry.The expression of NF-κB,LC3,and Beclin1 was examined by Western blotting.【Results】 The proliferation of SGC7901 cells was significantly inhibited by curcumin in a dose-dependent manner.After 40 μmol/L of curcumin treatment for 12h and 24 h,MDC and Hoechst 33342 staining were performed,and the autophagy in SGC7901 cells was increased compared to the control group in 12 h but the autophagy was decreased in 24 h.Then we performed Hoechst 33342 staining after 40μmol/L of curcumin treatment under the same conditions.The results showed that autophagosomes were increased in a time-dependent manner compared to the control group and the results of FCM indicated that the apoptosis rate was increased significantly after 40μmol/L of curcumin treatment for 24 h.In the course of autophagic apoptosis induced by curcumin,the autophagic stage was prior to executional stage of apoptosis.Western blot analysis showed that the expression of apoptosis related protein NF-κB,autophagy related proteins LC3 and Beclin1 was all up-regulated.【Conclusion】 Curcumin could significantly suppress the proliferation of SGC7901 cells through induction of autophagic apoptosis and the up-regulation expression of NF-κB,LC3 and Beclin1protein might be involved in the curcumin-induced autophagic apoptosis.
