活血消异方对子宫内膜异位症大鼠ICAM-1、MMP-9及VEGF的影响  被引量:22

摘  要:目的观察中药活血消异方对子宫内膜异位症(EMs)大鼠细胞间黏附分子-1(ICAM-1)、基质金属蛋白酶-9(MMP-9)、血管内皮生长因子(VEGF)表达水平的影响,探讨活血消异方抑制EMs发生的作用机理。方法建立大鼠EMs模型,随机分为活血消异方组、孕三烯酮组、模型对照组、正常对照组,采用免疫组织化学染色法检测给药3周及停药3周时各组大鼠子宫内膜组织中ICAM-1、MMP-9、VEGF的表达水平的变化。结果用药3周时,ICAM-1、MMP-9在模型对照组异位内膜中表达水平均高于其他各组(P<0.05),在活血消异方组异位内膜中的表达水平均低于孕三烯酮组(P<0.05)。VEGF在模型对照组异位内膜中表达最强,而孕三烯酮组中VEGF的表达较之模型对照组,差异无统计学意义(P>0.05),较之活血消异方组则表达增强(P<0.05)。停药3周后,ICAM-1、MMP-9及VEGF在活血消异方组异位内膜中的表达均低于模型对照组(P<0.05)。结论 "黏附、侵袭、血管生成的过程"(即"AAA通路")与EMs的发生密切相关,活血消异方可下调"AAA通路"相关因子的表达,通过抑制异位内膜的黏附、侵袭和局部新生血管的形成,降低异位内膜组织种植、侵袭、转移能力,最终抑制EMs的形成。Objective To observe tile influences of Huoxue Xiaoyi Fang on the expressions of intercellular adhesion molecule-1 (ICAM-1), matrix metalloproteinase-9 (MMP-9) and vascular endothelial growth factor (VEGF) in rats with endometriosis (EM) , and discuss the mechanism. Methods The model of EM was established in rats and the~ they were randomly divided into Huoxue Xiaoyi Fang group, gestrinone group, model group and normal group. The changes of expressions of ICAM-I , MMP-9 and VEGF were detected by using immunohistochemistry staining technique in all groups after giving mcdicinals for three weeks and after stopping medicinal administration for three weeks. Results After medicinal administration for three weeks, the expressions of ICAM-1 and MMP-9 were higher in model group than those in other groups ( P 〈 0.05 ) , and lower in Huoxue Xiaoyi Fang group than those in gestrinone statistical group (P 〈 0.05 ). The expression of VEGF was the highest in model group, and had no significance in gestrinone group compared with model gomp (P 〉 O. 05 ), and increased compared with Huoxue Xiaoyi Fang group (P 〈 0.05 ). The expressions of ICAM-1, MMP-9 and VEGF were all lower in Huoxue Xiaoyi Fang group than those in model group ( P 〈 0.05 ) after stopping medicinal administration for three weeks. Conclusion There is a close relation between the occurrence of EM and course of adhesion-aggression-angiogenesis (AAA pathway). Huoxue Xiaoyi Fang can down- regulate the expression of relative factors of AAA pathway, and preventing EM through inhibiting the adhesion, aggression and local angiogenesis and reducing the ability of planting, invading and transferring of endometriotic tissues.
