摘  要:目的:观察线粒体融合蛋白2(Mitofusin2,Mfn2)对昆明小鼠受精卵发育的影响,探讨其在早期胚胎发育中的重要作用。方法:采用荧光定量PCR对昆明小鼠Mfn2基因反义序列进行沉默有效性的筛选,将所筛选的序列转染入小鼠2细胞受精卵中,观察并统计囊胚形成速率及质量。结果:成功筛选得到有效Mfn2基因反义序列,Mfn2基因沉默后囊胚形成速度减慢,且正常囊胚形成率明显减低。结论:Mfn2基因沉默组中的囊胚形成速率和数量明显降低,提示Mfn2参与小鼠受精卵的发育,推测Mfn2的正常表达对小鼠植入前胚胎发育具有重要作用。Objective:To observe the effect of mitofusin 2(Mfn2) on zygote development of Kunming mice,and explore its important role in early embryonic development.Methods:Fluorescent quantitation PCR was used to screen the effective antisense sequence of Mfn2 gene in Kunming mice,and then the screening sequence was transfected into two-cell zygote,the blastulation rate and quality were observed statistically.Results:Effective Mfn2 gene antisense sequence was screened out successfully,the blastulation rate was slowed after Mfn2 gene silencing,and the formation rate of normal blastosphere was significantly reduced.Conclusion:The blastulation rate and quality were reduced significantly in Mfn2 gene silencing group,which indicates that Mfn2 participates in the process of zygote development in mice,and the normal expression of Mfn2 is important for preimplantation embryo development in mice.
