摘 要:核苷酸切除修复系统在维持基因组稳定、修复致癌因素所致的损伤及抗癌过程中发挥着重要作用。研究证实基因多态性可能引起氨基酸改变,然后导致表达的蛋白质异常,进而影响DNA修复能力。因此,基因多态性与肿瘤的发病风险和遗传易患性密切相关。同时在化疗过程中,其与肿瘤敏感性、中位生存时间等显著相关。切除修复交叉互补基因2(ERCC2)在核苷酸切除修复系统中发挥着重要作用,现就ERCC2基因多态性与胃癌的易患性以及化疗药物之间的关系予以综述。Nueleotide excision repair system plays a pivotal role in maintaining integrity of genome and repai- ring the damage caused by carcinogenic factors. Published studies showed that changing of amino acid caused by polymorphism may convert the structure or function of the proteins, and then lead to the alteration of DNA repair capacity. Consequently there is a close relationship between genetic polymorphisms and susceptibility and genetic predisposition of cancers. In chemotherapy of cancers, genetic polymorphisms are connected with tumor sensitivity and the median survival time as well. ERCC2 is a major member in nucleotide excision repair pathway,here is to make a review on the association between polymorphism of ERCC2 and susceptibility and chemotherapy drugs of gastric cancer.
