LRRK2基因核心启动子的预测

摘　　要：目的运用生物信息学方法对LRRK2基因的启动子区域进行预测,以指导LRRK2基因核心启动子的克隆,为LRRK2基因的转录调控机制的研究奠定基础。方法在数据库中查询LRRK2基因相关的序列信息,运用多种在线软件对LRRK2基因的转录起始位点、第一个外显子和启动子进行预测,并对预测结果进行综合分析。结果将LRRK2基因脑组织主要剪切本(ENST00000298910,相当于NM_198578)的启动子定位于-803～-1区间。结论 LRRK2基因主要剪切本(ENST00000298910)的启动子区间可能定位于-803～-1区间。Objective To predict the promoter region of leucine-rich repeat kinase 2 （LRRK2） gene with bioinformatics, and to direct the core promoter＇s clone of LRRK2 gene for the basis of research on the transcription regulation mechanism of LRRK2 gene. Methods The information on LRRK2 gene-related sequence was searched in the database,and several on-line softwares were used to predict the transcriptional start site ,the first exon,and the promoter regions of LRRK2 gene. Comprehensive analysis of the above predicted result was performed. Results ENST0000098910 （ corresponding to NM_198578） ,the major transcript in the brain,was the target transcript. The basal promoter of LRRK2 was predicted to be located at - 803/- 1 region. Conclusion The basal promoter of LRRK2 may be located at - 803/- 1 region.
