摘 要:目的探讨片仔癀对局灶性脑梗死大鼠大脑皮层水通道蛋白4(AQP4)表达的影响及其分子作用机制。方法制备MCAO大鼠模型,将SD大鼠随机分为7组:假手术组,模型组,片仔癀大、中、小剂量组,尼莫地平组和脑得生组。造模前给药3d,造模后给药至第7天取材,采用免疫组织化学法观察各组大鼠大脑皮层AQP4的表达情况。结果与假手术组相比,各造模组AQP4的表达水平显著升高(P<0.05);与模型组比较,各给药组AQP4的表达均显著下降(P<0.05);与尼莫地平组和脑得生组相比,片仔癀各剂量组AQP4的表达水平有所下降(P<0.05)。结论片仔癀可减少局灶性脑梗死大鼠大脑皮层AQP4的表达,减轻脑水肿,从而起到脑保护的作用。Objective To explore the effect of Pien Tze Huang(PTH) on aquaporin-4 (AQP4) expression in focal cerebral infarction rats. Methods The Sprague - Dawley (SD) rat models were prepared by middle cerebral artery embolization (MCAO). Seventy SD rats were randomly divided into seven groups:Sham operation group, model group, high dose group of PTH, middle dose group of PTH,small dose group of PTH, nimodipine group,and naodesheng group. Cortex AQP4 expression were observed by immunohisto chemistry method. Results Compared with the sham operation group, expression were increased in other group (P〈0.05). Corn pared with the model group, the AQP4 expression in treatment group were significantly decreased (P〈0.05). Compared with nimo- dipine group and naodesheng group,AQP4 expression PTH groups were decreased (P〈0.05). Conclusion PTH could reduce the AQP4 expression of the cerebral cortex in focal cerebral infarction rats, and protect the function of the brain.
