摘 要:目的探讨禁食条件下急性细菌脂多糖(LPS)暴露对小鼠血糖的影响。方法雄性成年ICR健康小鼠随机分为对照组和LPS组,LPS组单次腹腔注射2 mg/kg LPS,对照组单次腹腔注射相同体积的生理盐水。注射12 h后剖杀小鼠,取血清检查生化指标;用实时定量PCR检测肝脏糖代谢相关基因葡萄糖-6-磷酸酶(G-6-Pase)和磷酸烯醇式丙酮酸羧化酶(PEPCK)mRNA水平。结果 LPS注射后24 h内血糖水平持续降低;葡萄糖耐量(GTT)实验显示,LPS引起小鼠对葡萄糖耐量降低;胰岛素耐量(ITT)实验显示,LPS引起小鼠胰岛素敏感性降低;实时定量PCR结果显示,单次给予LPS显著下调g-6-pase、pepck mRNA水平。结论单次LPS抑制肝脏糖异生,导致持续性低血糖。Objective To investigate the effect of acute lipopolysaccharide (LPS) exposure on the serum glucose in mice under the condition of fasting. Methods Male ICR mice were randomly divided into two groups. In the LPS group, mice were intraperitoneally injected with a single dose of LPS(2 mg/kg). In control group, mice were injected with saline. Mice were sacrificed 12 h after LPS. Blood serum was collected for the measurement of bio-chemical parameters. The expressions of hepatic gtucose-6-phosphatase and phosphoenolpyruvate carboxykinase were determined using real-time PCR detecting system. Results The levels of serum glucose were persistently decreased in LPS-treated mice, GTT results showed that glucose tolerance decreased in LPS-administered mice. However, ITF results showed that insulin tolerance increased in LPS-administered mice. LPS significantly inhibited the expression of hepatic glucose-6-phosphatase and phosphoenolpyruvate carboxykinase mRNA. Conclusion Acute lipopolysaccharide exposure induces persistent hypoglycemia through decreasing hepatic gluconeogenesis.
