摘  要:目的考察Tecra^(TM)大肠杆菌O157酶联免疫快速方法的准确度和特异度,与经典培养法进行比较,并针对不同种类的食品样品进行应用评价。方法用不同来源的大肠杆菌O157菌株考察其准确度;用多种标准非大肠杆菌O157菌株考察其特异度;利用加标回收实验结合国标方法测定其检出限;用Tecra^(TM)大肠杆菌O157酶联免疫快速方法与国标中经典法和免疫磁珠法对不同种类的食品样品进行检测,比较并评价其应用效果。结果该方法准确度为100%;特异度为100%;检出限为1 CFU/25 g;630份实测样品的结果与国标方法对比其灵敏度为100%。结论该方法快速、简便、高效和准确,适宜食源性突发公共卫生事件及大批量样品的大肠杆菌O157快速检测。OBJECTIVE To examine and compare the accuracy and specificity of the TecraTM E.coli O157 enzyme-linked immunosorbent rapid method with classical methods, evaluate the application for different types of food samples. METHODS Accuracy of the TectaTM E. eoli O157 enzyme-linked immunosorbent rapid method is investigated with different sources of E. eoli O157 strains, its specificity is examined using the standard strains of a variety of non-E, coli 0157, its detection limit was measured by the test of recovery experiments combined with national standard methods, application of this method was compared and evaluated by detecting different food samples with the TeeraTM E. eoli O157 enzyme-linked immunosorbent rapid method and national standard methods (classical methods and immunomagnetic bead separation). RESULTS Accuracy, specificity and detection limit of this rapid method is 100%, 100% and 1 CFU/25 g respectively. Compared with national standard methods, Sensitivity of the TecraTM E. coli O157 enzyme-linked immunosorbent rapid method is 100% by detecting 630 samples. CONCLUSION This method is rapid, simple, efficient and accurate, and suitable for rapid detection of E. coli O157 in food-borne public health emergencies and large amount samples.
