摘 要:目的:研究中药制剂参芪扶正注射液含药血清对胃癌MGC-803细胞侵袭能力及Tenascin-C(Tn-C,腱糖蛋白-C或韧粘素-C)mRNA表达的影响。方法:分别将高浓度(0.16g/ml)、中浓度(0.8g/ml)、低浓度(0.4g/ml)不同剂量的中药制剂参芪扶正注射液注射于小鼠制备含药血清及对照血清。设立空白组,对照血清组,低、中、高浓度参芪扶正注射液组,对胃癌MGC-803细胞株培养48小时,采用MatrigelTM方法观察各组细胞株的侵袭情况。然后分别用不同剂量含药血清培养胃癌细胞,采用RT-PCR法检测各组胃癌细胞中Tn-C表达的影响。结果:与空白对照组比较,参芪扶正注射液能明显抑制胃癌细胞侵袭能力,且浓度越高抑制作用越强,高浓度含药血清使细胞的侵袭能力下降34.7%。中药制剂参芪扶正注射液高、中、低剂量组均能明显降低Tn-C值,且降低程度与含药血清浓度正相关。结论:中药制剂参芪扶正注射液能明显抑制胃癌MGC-803细胞株体外侵袭能力,并且下调胃癌细胞中Tn-C的表达,这可能是其抑制肿瘤侵袭的分子机制。Objective:To investigate the effects of Shenqi Fuzheng injection on the invasiveness of cells and expressions of Tenascin - C ( Tn - C) in human gastric cancer MGC - 803 cells. Methods: MGC - 803 gastric cancer were cultured in medium for 48 hours that taken from rats, which injected different density of Shenqi Fuzheng injection (0.04g/m1,0.08g/ml and O. 16g/ml) for 1 week. MatrigelTM test was performed to detect invasion ability of cells, and RT - PCR was used to check Tn - C mRNA expression. Results: In treated groups, invasiveness of cells was most obviously( the inhibit ratio was 34.7% in 0.16g/ml group). The expressions of Tenascin - C were lower than those of the control. There was significant difference between the high concentration group and the control. Conclusion : The expressions of Tenascin - C are down - regulated when gastric cancer MGC - 803 cells were treated with Shenqi Fuzheng injection at high concentration, therefore the invasiveness of gastric tumors maybe reduced.
