摘 要:目的研究家蝇胚胎细胞MDEC-07114抗菌蛋白对人髓样白血病细胞K562的抑制作用。方法利用脂多糖诱导家蝇胚胎细胞MDEC-07114,提取抗菌蛋白,配制成40、80、160、320和640μg/ml 5个实验组,同时以人脐静脉血管内皮细胞作为正常对照组。采用MTT法测定不同浓度抗菌蛋白对人髓样白血病细胞K562和人脐静脉血管内皮细胞的抑制作用,流式细胞术观察5种浓度抗菌蛋白对人髓样白血病细胞K562细胞周期及凋亡的影响。结果①MTT显示5种浓度抗菌蛋白对K562细胞均有明显的抑制作用,而对人脐静脉血管内皮细胞无抑制作用。②流式细胞术显示对K562细胞S期的影响与阴性对照组相比差异有统计学意义(P<0.05)。各浓度组均可引起K562细胞的G0/G1期百分比上升,细胞增殖指数PI降低。与阴性对照组相比,各浓度组引起的K562细胞细胞凋亡率差异无统计学意义(P>0.05)。结论家蝇胚胎细胞抗菌蛋白对肿瘤细胞具有明显的抑制作用,而对人体正常细胞无抑制作用。家蝇胚胎细胞抗菌蛋白可阻滞K562细胞周期的正常发育。Objective To research the inhibitory action of the antibacterial protein formydoid leukemia K562. Methods The logarithmic growth phase' s embryonic cells of the methods domestica were induced by LPS. The antibacterial protein was extracted and was prepared 40,80,160,320 and 640trg/ml 5 density groups, the human umbilical vein vascular endothelial cells were been normal control group. The inhibitory action on the human myeloid leukemia cells K562 and the human umbilcali vein vascular endothelial cells were made by MTT experiments. The cell cycle and the cell apoptosis ratio of K562 cells were detected in five density groups of antibacterial protein by flow cytometry. Results The MTT examination demonstrated that the five density antibacterial proteins had all inhibition activities on K562 cells,and had no inhibition activities on human umbilical vein vascular endothelial cell. For K562 cells, the influence of different density antibacterical peptides to S phase had the significance differ ence, compared with the negative group( P 〈 0. 05). Each density groups produced all rise of the G0/G1 of K562 cells and reduce of the cell multiplication index PI. Compared with the negative control group, the apoptosis ratio in various density groups had not significant difference( P 〉 0. 05 ). Conclusion The antibacterial protein from the embryonic cell MDEC-07114 of the Musca dom- estica has significance inhibitory activity on tumor cells, and has no inhibition activity on normal human cells. The antibacterical peptide from the embryonic cells of the musca domestica maybe arrest normal development of the K562 cells cycle.
