摘  要:在夏季强光照条件下进行遮荫处理,研究光照诱导型新梢白化茶树品种"黄金芽"叶片基因表达谱的差异。结果显示,自然强光照(无遮荫)和遮荫处理reads数分别为5557292个和6309925个,参考基因Unigene数79797个。无遮荫和遮荫处理之间表达丰度明显差异的基因1392个,其中遮荫引起表达上调的基因902个,表达下调的基因490个;214个基因在无遮荫条件下表达完全受到抑制,72个基因在遮荫处理表达完全受到抑制。受强光照抑制的基因包括:光诱导蛋白、肽链内切酶抑制蛋白、蔗糖和氨基酸转运因子、核糖体失活、糖结合蛋白等基因;但遮荫抑制了查耳酮合成酶基因3(CHS3)表达。据此推断,"黄金芽"茶树光诱导蛋白表达受到强光条件抑制,可能是其对强光环境压力的耐受较弱,进而产生新梢白化的原因之一。研究还表明,"黄金芽"茶树品种的基因组成与"福鼎大白茶"之间存在1/3左右的碱基变异。Cene expression profiles in shaded and unshaded leaves of tea cuhivar ' Huangjinya' , a light-induced albino tea cultivar, were analyzed. Total reads in the expression profile was 5557292 in unshaded leaf and 6309925 in shaded leaf, with total 79797 unigene. 1392 genes showed significant difference in expression level, among which 902 genes were up-regulated and 490 down-regulated in shaded leaf. 214 genes were completely inhibited in unshaded leaf and 72 genes were completely inhibited by shading. The genes which were inhibited by strong sunlight involved in genes of light- inducible protein CPRF2, endopeptidase inhibitor, transporter of amino acid and sucrose, type 2 ribosome-inactivating protein and sugar binding protein. Expression of chalcone synthase gene 3 (CHS3) was inhibited by sunlight shading. The inhibition of light-inducible protein CPRF2 expression by strong sunlight suggests that the tolerance to strong light stress was weak in cultivar ' Huangjinya' and this resulted in its albinism. The results also shows that there were about 1/3 difference in base site between cultivars ' Huangjinya' and ' fudingdabai'
