摘 要:目的探讨红景天苷(SD)诱导大鼠骨髓间充质干细胞(BMSCs)向多巴胺能神经细胞分化的影响。方法实验分为红景天苷组,维甲酸(RA)组和空白对照组,SD和RA分别诱导BMSCs 1d、3 d、6 d和9 d,细胞免疫荧光化学法检测神经细胞相关标志分子神经元特异性烯醇化酶(NSE)、神经微管相关蛋白2(MAP2)、抗微管蛋白(β-TubulinⅢ)和神经胶质原纤维酸性蛋白质(GFAP)以及与多巴胺能神经元相关的关键酶多巴脱羧酶(DBH)和多巴胺-β-羟化酶(DDC)的表达;逆转录-聚合酶链反应(RT-PCR)检测诱导前后神经元特异性烯醇化酶和神经胶质纤维酸性蛋白、核受体相关因子1(Nurr1)、神经营养因子-3(NT-3)和酪氨酸羟化酶(TH)mRNA的表达;酶联免疫吸附法评价诱导前后细胞表达NT-3、脑源性神经营养因子(BDNF)的水平。结果 SD和RA分别诱导BMSCs 6 d和9 d时细胞增殖明显增强,与空白对照组相比差异有统计学意义(P<0.05),诱导3 d后NSE、MAP2和β-TublinⅢ表达阳性,红景天苷组GFAP表达阳性,维甲酸组GFAP表达阳性。6 d时NSE表达丰度上调;1 d、3 d和6 d诱导组Nurr1 mRNA表达丰度上调,红景天苷组NT-3 mRNA表达水平与维甲酸组比较明显上调,SD组6 d时TH mRNA表达上调。诱导组DBH和DDC阳性率与对照组相比差异具有统计学意义(P<0.05)。诱导组NT-3和BDNF细胞因子含量增加与对照组相比差异具有统计学意义(P<0.01)。结论红景天苷能诱导BMSCs分化为具有多巴胺功能的神经细胞。Objective To investigate the effect of salidmside (SD) on rat bone marrow mese.nehymal stem cells (BMSCs) differentiating into the dopaminergic nerve cells. Methods Experiment groups were divided into salidroside (SD) group, retinoic acid (RA) group, and the control group. BMSCs were induced at 1 d, 3 d, 6 d and 9 d by salidroside and retinoic acid, respectively. The expressions of relevant marker molecules of nerve cells, such as neuron-specific enolase (NSE), microtubule-associated protein 2 (MAP2), β-Tubhn III, glial fibfillary acidic protein (GFAP), and the key enzyme of dopaminergic neuron dpa decarboxylase (DDC) and dpamine-beta-hydmxy (DBH) were detected by imrramofluorescenee. Expressions of NSE, nuclear receptor-related factor 1 (Nurrl), GFAP, neurotrophin 3 ( NT-3 ) and trosine hydroxylase (TH) mRNA were detected by reverse transcription polymerase chain reaction (RT-PCR). The levels of NT-3 and brain-derived neurotrophic factor (BDNF) was examined by enzyme-linked irrantmosorbent assay. Results The cellular proliferation was increased significantly at 6 d and 9 d in salidroside group and RA group than that of control group (P 〉0.05). The positive expressions of NSE, MAP2, and β-Tublin m were observed after induced for 3 d, whereas GFAP expression was negative in salidmside group. NSE mRNA expression was up-regulated at 6 d and the level of Nurrl mRNA was also up-regulated at 1 d, 3 d, and 6 d in the induced groups. Expressions of NT-3 mRNA were increased significantly at salidroside group compared to retinoic groups and trosine hydroxylase mRNA expression was up-regulated at 6 d in SD induced groups. The positive ratios of dpa decarboxylase and dpamine-beta-hydroxy were higher in the induced groups comgared with control group (P 〈0.05). There is significant difference in cytokine level of NT-3 and BDNF between the induced groups and control group (P 〈0.01). Conclusion Salidroside can induce BMSCs differeraiating into depaminerg
