秦艽种子蛋白粗提取研究

摘　　要：秦艽种子蛋白粗提取是电泳法快速检测秦艽种子质量的前提。本实验采用正交试验方法,对提取过程中的料液比、缓冲液类型与pH、浸提时间、浸提液、PVP用量、SDS等主要影响因素进行了研究。初步确定在低温条件(4℃)下,秦艽种子蛋白提取较佳条件为:料液比1∶10,pH值8.0的0.05 mol/L Tris-HCl(1%DTT)缓冲液,浸提时间1 h;并且在这种条件下秦艽种子提取的谷蛋白最多。Gentiana macrophylla Pall. seeds protein coarse extraction is the preliminary for the G. macrophylla Pall. seeds quality dectection with electrophoresis method. This experiment used the orthogonal method. All the factors including the ratio of sample to solution, buffers and pH value, time, media, amount of PVP and SDS which might affect the extraction of proteins from G. macrophflla Pall. seeds were studied. The experiment produced the best result in the condition for 1.0 h in 0.05 mol/L Tris-HC1 （ 1% DTT） buffer with a 1 ： 10 ratio of sample to solution and pH 8.0 at low temperature conditions （4 ℃ ）.
