摘  要:目的观察补阳还五汤对血管性痴呆(Vascular Dementia,VD)大鼠海马CA1区突触传递LTP效应及对海马组织ERK2与CaMKIIβ基因表达的影响。方法采用四血管阻断法建立大鼠VD动物模型。术后25d行水迷宫检测,30d采用在体大鼠海马CA3侧枝-CA1区长时程增强(LTP)记录方法记录海马CA1区LTP。采用RT-PCR方法检测大鼠海马CA1区脑组织ERK2与CaMKIIβmRNA表达。结果与模型组比较,补阳还五汤和尼莫地平组水迷宫实验平台象限游泳时间/游泳总时间、平台象限游泳距离/游泳总距离的比值显著增加,LTP检测实验中兴奋性突触后电位(excitatory postsynaptic potential,EPSP)斜率显著上升,大鼠海马ERK2与CaMKIIβmRNA表达显著增加。补阳还五汤与尼莫地平组相比,上述各指标均无显著差异。结论补阳还五汤可显著改善血管性痴呆大鼠学习记忆障碍,其作用可能与增强海马CAl区LTP,上调ERK2与CaMKIIβmRNA表达有关。Objective To investigate the effects of BYHWD on the LTP of synaptic transmission in hippocampal CA3 region and the variability of mRNA expressions of ERK2 and CaMKⅡβ in VD rats. To explore the mechanism of BYH- WD on improving learning and memory. Methods Rats were randomly divided into 4 groups : Sham operation group, VD + BYHWD group, VD + nimodipine group and VD group. The rats of VD + BYHWD group, VD + nimodipine group and VD group were made by four-vessel occlusion. The rats of VD + BYHWD group and VD + nimodipine group were delivered- cured by BYHWD and nimodipine by intragastric administration after awakening for 30days. Morris water maze tests were used to observe the learning and memory ability. The change of synaptic plasticity in hippocampus CA1 region of rats were detected, HFS was given by CA3 Schaffer collateral-commissural pathway, and then LTP of hippocampal CA1 region was re- corded. RT-PCR was used to observe ERK2 and CaMKⅡβ mRNA expression in the brain tissue. Results Water maze de- tection : The percent of distance and time between the quadrant previously containing the platform was shortened significantly (P 〈 0. 05). The escape latency of rats of VD + BYHWD group and VD + nimodipine group were significantly shorter as compared with Model group, and the percent of distance and time in the quadrant previously containing the platform was in- creased significantly (P 〈 0.05). LTP detection:Compared with Sham operation group,the slope of EPSP of VD group had decreased significantly ( P 〈 0. 05 ). Compared with VD group, the slope of EPSP of rats inof VD + BYHWD group and VD + nimodipine group had raised significantly( P 〈 0.05 ). And compared between VD + BYHWD group and VD + nimodipine group there washad no significant deviation ( P 〉 0.05 ). RT-PCR detection: Compared with Sham operation group, ERK2 and CaMKⅡβ mRNA expression of VD group had decreased significantly(P 〈 0.05 ), Compared with VD group,ERK2 and CaMKⅡβ
