摘 要:目的研制化脓性脑膜炎常见细菌检测基因芯片,为化脓性脑膜炎病原学明确诊断提供新技术。方法在设计化脓性脑膜炎常见细菌:脑膜炎奈瑟菌、肺炎链球菌、流感嗜血杆菌、金黄色葡萄球菌、表皮葡萄球菌、大肠埃希菌、其他凝固酶阴性葡萄球菌的特异性探针的基础上,通过芯片基片的制备、点样、固定、样品扩增标记、杂交及扫描等工艺过程的优化,完成基因芯片研制。结果初步研制出常见化脓性脑膜炎病原菌检测的基因芯片。结论研究的基因芯片能够用于化脓性脑膜炎常见病原菌的诊断和分型检测,有望成为一种化脓性脑膜炎脑脊液常见病原菌的高效检测技术。Objective To develop a gene chip for purulent meningitis common pathogens detection, so as to provide new technology to con- firm the diagnosis of pttrulent meningitis etiology. Methods Specific probes were designed for common pathogens of purulent meningitis in- cluding Neisseria meningitidis, Streptococcus pneumoniae, Haemophilus influenzae, Staphyloeoceus aureus, Staphylococcus epidermidis, Es- chetichia coli,and coagulase-negative staphylococci;the processes included preparation of the chip substrate,ehip spotting and immobiliza- tion, sample amplification and labeling, hybridization and chip scanning. Results The processes were optimized, and the common purulent meningitis pathogen detection gene chip was initially developed. Conclusion The developed gene chip can be used for diagnosis and iden- tification of common pathogens of purulent meningitis, which is expected to he a highly effective meningitis pathogen detection technology.
