摘  要:目的观察17-AAG对人乳腺癌MCF-7细胞株STAT3、VEGFmRNA及蛋白表达水平的影响。方法体外培养MCF-7细胞,分别给予不同剂量及不同作用时间的17-AAG,采用四甲基偶氮唑盐微量酶反应比色法(MTT法)检测细胞增殖能力;RT-PCR和Westernblot法检测各组细胞STAT3和VEGFmRNA及蛋白的表达水平。结果0.165~10.000mg/L17-AAG作用24h、48h对MCF-7细胞有显著的抑制作用,且有明显的时间、剂量依赖性;1.0、2.0、3.0、5.0mg/L17.AAG作用48h后,各组细胞STAT3和VEGFmRNA及蛋白表达水平均下调,且具有浓度依赖性。结论17一AAG对人乳腺癌MCF.7细胞的增殖具有明显的抑制作用,并能抑制STAT3和VEGFmRNA及蛋白的表达,且具有剂量依赖性。Objective To investigate the effects of 17-AAG on VEGF and STAT3 expressions in human breast cancer MCF-7 cells. Methods MCF-7 cells were cultured in DMEM roediuro;MTr method was performed to evaluate the inhibitory effects of 17-AAG with dif- ferent time and different doses on proliferation of MCF-7 cells;the genes and proteins expressions of VEGF and STAT3 were determined by RT-PCR and Western blot analysis. Results With treatment of 0.165-10.000 mg/L 17-AAG for 24 hours and 48 hours, significantly inhibi- tion of the MCF-7 cells proliferation was observed in both time- and dose-dependent manner. The assay of RT-PCR and Western blot showed that the genes and proteins expressions of VEGF and STAT3 in MCF-7 cells induced by 17-AAG were significantly down-regulated in a dose-dependent manner when the cells were treated with 1.0,2.0,3.0,5.0 mg/L 17-AAG for 48 hours. Conclusion 17-AAG can in- hibit the cell proliferation and down regulate the genes and proteins expressions of VEGF and STAT3 of human breast cancer MCF-7 cells in vitro.
