摘  要:目的探讨胰岛素对血管内皮细胞中miRNA-21的表达和分泌影响。方法用不同浓度胰岛素(0、1、10、50、100和1000nmol/L)干预人脐静脉内皮细胞(HUVECs),48h后收集细胞上清液及细胞,用实时荧光定量PCR方法检测细胞及上清液中miRNA-21的表达情况。结果与0nmol/L胰岛素组相比,10、50、100和1000nmol/L胰岛素组HUVECs中miRNA-21表达均下调(P均<0.05),1000nmol/L胰岛素组细胞中miRNA-21表达最低(P<0.05),但1与0nmol/L胰岛素组HUVECs中miRNA-21表达差异无统计学意义(P>0.05)。与0nmol/L胰岛素组相比,1和10nmol/L胰岛素组HUVECs上清液中miRNA-21表达升高(P<0.05),而50、100和1000nmol/L胰岛素组上清液中miR-NA-21表达降低(P均<0.05),50nmol/L胰岛素组上清液中miRNA-21表达含量最低(P<0.05)。结论不同浓度胰岛素可影响HUVECs中miRNA-21表达,这种变化反映了不同浓度胰岛素对HU-VECs的凋亡和增殖影响。Objective To explore the expression and secretion of microRNA-21 (miRNA-21) in vascular endothelial cells treated with insulin. Methods Human umbilical vein endothelial cells (HUVECs) were treated with different concentrations of insulin (0, 1, 10, 50,100 and 1000 nmol/L respectively) for 48 hours, and then HUVECs and supernate were collected to detect miRNA-21 expression with Real-time PCR assay. Results The expression of miRNA-21 was down-regulated in HUVECs with 10, 50, 100 and 1000 nmol/L insulin groups respectively (P all〈0. 05) and was reduced most significantly in 1000 nmol/L insulin group (P〈0. 05), but there was no difference in miRNA-21 expression in 0 umol/L and 1 nmol/L insulin groups(P〈0. 05). The expressions of miRNA-21 were up- regulated significantly in supernate of the 1 umol/L and 10 umol/L insulin groups (P〈0. 05), however, the expressions of miRNA-21 were down-regulated in supernate of 50, 100 and 1000 nmol/L insulin groups respectively (P 〈 0. 05). Conclusion Different concentrations of insulin could affect the expression of miRNA-21 in HUVECs, which possibly reflect the apoptosis and proliferation of HUVECs induced by different concentrations of insulin.
