摘  要:目的:检测β-连环素(β-catenin)在宫颈鳞癌组织中的表达情况,初步探讨其与肿瘤发生、发展及转移的生物学关系。方法:收集32例正常宫颈组织、34例宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)及宫颈鳞癌(squa-mous cell carxinoma,SCC)组织86例,采用RT-PCR技术和免疫组化SP法检测组织中β-catenin mRNA及蛋白的表达情况。结果:β-catenin mRNA在正常宫颈组织中的相对表达系数为1.053±0.123,宫颈上皮内瘤变(CIN)组织为0.924±0.227,宫颈鳞癌组织为0.806±0.05,三者比较差异具有统计学意义(P<0.05);β-catenin蛋白在正常宫颈组织、宫颈上皮内瘤变组织和宫颈鳞癌组织中的阳性表达率分别为93.75%(30/32)、61.76%(21/34)和44.19%(38/86)。而且β-catenin的表达与组织分化程度、有无淋巴结转移和临床分期关系密切(P<0.05),而与患者年龄和组织类型无关(P>0.05)。结论:β-catenin基因在宫颈癌中表达增高,且与肿瘤分化程度、有无淋巴结转移和临床分期相关,提示其高表达可能参与肿瘤的发生、发展,检测β-catenin有助于了解宫颈癌生物学行为和预后。Objective: To detect the expression of 13 - catenin in cervical squamous cell carcinoma ( SCC), and explore the biolog- ical relationship between β - catenin and oncogenesis, development, and metastasis of cervical SCC. Methods: Thirty - two normal cervical specimens, thirty - four cervical intraepithelial neoplasia (CIN) specimens, and eighty - six cervical SCC specimens were collected, RT - PCR and immunohistochemical SP method were used to detect the expression levels of β - catenin mRNA and protein. Results: The relative expression coefficient of β - catenin mRNA in normal cervix, CIN, and cervical SCC were ( 1. 053± 0. 123 ), ( 0. 924± 0. 227 ), and (0. 806 ± 0. 05 ) , respectively, there was statistically significant difference among the three groups ( P 〈 0. 05 ) ; the positive expression rates of β - catenin protein in normal cervix, CIN, and cervical SCC were 93.75% (30/32) , 61.76% (21/34) , and 44. 19% (38/86) , respectively. The expression of β - catenin was closely correlated with degree of histological differentiation, lymph node metastasis, and clinical stages ( P 〈 0. 05 ), but the expression of β - catenin was not related to age and histological types ( P 〉 0. 05 ) . Conclusion : The ex- pression of β - catenin increases in cervical SCC, and it is correlated with degree of histological differentiation, lymph node metastasis, and clinical stages, which indicates that β - catenin may participate in oncogenesis and development of cervical SCC, detecting β - catenin is helpful to understand biological behavior and prognosis of cervical SCC.
