摘  要:目的探讨黄芪皂苷体外抗神经胶质瘤活性及相关机制。方法采用体外培养的SHG44细胞株为研究对象,流式细胞仪检测黄芪皂苷作用后SHG44细胞凋亡率,免疫印迹法检测细胞凋亡关键蛋白p53、Bax、Bcl-2的表达变化。结果黄芪皂苷可诱导SHG44肿瘤细胞凋亡;黄芪皂苷可引发SHG44肿瘤细胞促凋亡蛋白p53、Bax高表达,抗凋亡蛋白Bcl-2表达降低。结论黄芪皂苷可通过p53信号通路激活和Bcl-2家族介导的细胞色素C途径诱导癌细胞凋亡,从而对神经胶质瘤细胞产生抑制作用。Objective To explore the anti-glioblastom activities in vitro and mechanisms of Astragalosides.Methods Apoptosis rate of SHG44 cell was valued by using flow cytometry(FCM) analysis.The expression level of p53,Bax and Bcl-2 was determined by western blotting.Results The FCM data showed that Astragalosides induced apoptosis of SHG44 cell.The expression level of p53 were significantly increased when treated with Astragalosides.The expression of Bax was also increased markedly,while Bcl-2 was reduced after treatment with the drug.Conclusion Our findings demonstrated that Astragalosides exhibits great anti-glioblastom activities on bladder carcer cells,possibly by triggering p53 signaling way and cytochrome C apoptotic pathway which mediates by Bcl-2 family.
