摘 要:目的研究蓝莓提取物对人结肠癌细胞株HCT116增殖的体外抑制作用及其机制。方法应用不同浓度梯度蓝莓提取物处理人结肠癌HCT116细胞,噻唑蓝(MTT)比色法检测肿瘤细胞增殖情况,应用流式细胞术检测蓝莓提取物对HCT116细胞体外凋亡情况影响,并进一步研究了Caspase-3活性以及NF-κB表达变化情况以探讨相关机制。结果蓝莓提取物对HCT116细胞的增殖抑制作用呈现明显的剂量及时间效应关系;流式细胞仪检测显示各浓度(0.25、0.50、1.00、2.00 mg/L)蓝莓提取物处理组HCT116细胞平均凋亡率分别为(8.86±1.03)%、(16.33±1.99)%、(49.29±6.73)%、(63.68±8.26)%,除0.25 mg/L组外,均显著高于对照组的(6.62±0.97)%;1.00 mg/L蓝莓提取物还可显著升高肿瘤细胞中Casepase-3活性,抑制NF-κB表达。结论蓝莓提取物可能通过激活Casepase-3级联反应通路和抑制NF-κB表达,诱导人结肠癌HCT116细胞凋亡从而抑制其体外的生长与增殖。Objective To illustrate the growth inhibition and apoptosis in human colon cancer HCT116 cells treated with blueberry extracts (BEs). Methods The antitumor activities of BEs against HCT116 cell were tested by MTT method, and flow cytometry was used to examine the apoptosis of cancer cells after treated with different concentration of BEs in vitro. The activity of Caspase-3 and NF-κB expression were also examined to explore the mechanisms. Re- suits Cell proliferation was inhibited after treated with BEs. Flow cytometry analysis revealed significantly increase in apoptosis rate of HCT116 cells (16.33±1.99)%, (49.29±6.73)%, (63.68±8.26)% contrast with control group [(6.62±0.97)%] after treatment with 0.50 mg/L, 1.00 mg/L or 2.00 mg/L BEs with exception of group of 0.25 μg/mL [(8.86±1.03)%]. The activity of Caspase-3 was signifieantly increased, while NF-κB was reduced when treated with BEs for 36h and 72h. Conclusion Our findings demonstrate that BEs exhibit great antitumor activities, possibly by triggering Caspase- 3 and NF-κB apoptotic pathway.
