摘 要:目的探讨小干扰RNA(siRNA)干扰非转移性黑色素瘤糖蛋白B(GPNMB)表达对人胶质母细胞瘤细胞系生物学特性的影响。方法将GPNMB siRNA片段通过脂质体转染人胶质母细胞瘤细胞系U251和SHG44,Western blot检测GPNMB蛋白表达水平;Annexin V/PI双染法检测GPNMB-siRNA对细胞凋亡的影响;MTT法检GPNMB-siRNA对细胞增殖的影响;Transwell法检测GPNMB-siRNA对细胞侵袭性的影响。结果 GPNMBsiRNA能有效抑制人胶质母细胞瘤细胞系U251和SHG44中GPNMB的表达,与空白对照组比较,GPNMB的缺失对细胞凋亡影响不大,可显著抑制细胞的增殖和侵袭(P<0.05)。结论 GPNMB基因敲除对人胶质母细胞瘤细胞系U251和SHG44的凋亡无显著影响,但可显著抑制其增殖力和侵袭力,表明GPNMB在人胶质母细胞瘤的发生、发展中起着重要作用,提示GPNMB可以作为治疗人胶质母细胞瘤的潜在靶点。Objective To explore the effect of siRNA-induced glycoprotein nonmetastatic melanoma protein B(GPNMB) silence on biological behavior in human glioblastoma cells.Methods The chemically synthesized GPNMB-siRNA was transfected into U251 and SHG44 cells by liposome.The expression levels of GPNMB protein were detected by western blot.The changes of cell apoptosis and proliferation were analyzed by double staining with Annexin V and propidium iodide and MTT colorimetry,separately.Cell invasion potential was evaluated by Transwell invasion assay.Results Transfection of GPNMB-siRNA to U251 and SHG44 cells significantly abolished GPNMB expression.Compared with blank control group,GPNMB-siRNA had no significant effect on cell apoptosis,but markedly inhibited the proliferation and invasion(P 0.05).Conclusion The siRNA-mediated silence of GPNMB can effectively inhibit cell proliferation and invasion in U251 and SHG44 cells,but has little effect on apoptosis.These results suggest that GPNMB play a vital role in the gliomagenesis and development of glioma,and may be used as a potential therapeutic target in human glioma.
