摘 要:目的观察慢性阻塞性肺疾病(COPD)大鼠不同时期肺组织中血管内皮生长因子(VEGF)mRNA表达水平的变化及肺血管超微结构的变化,并探讨二者的相关性。方法 48只清洁级健康雄性Wistar大鼠随机分为对照组24只和COPD模型组24只,COPD模型组又分为COPD 1、2、4周组各8只。采用气管内注入脂多糖及烟熏方法制作COPD模型。分别于第1、2、4周处死大鼠,观察不同时期COPD大鼠肺血管超微结构及肺组织中VEGF mRNA表达水平。结果随着COPD病情进展,电镜下肺血管的超微结构改变越明显,呈现阶梯状逐步加重的趋势,VEGF mRNA的表达也逐渐增高,COPD 1、2、4周组大鼠肺组织VEGF mRNA的表达均高于对照组(P<0.05,0.01);COPD 2周组大鼠肺组织VEGF mRNA的表达高于COPD 1周组(P<0.05);COPD 4周组大鼠肺组织VEGF mRNA的表达高于COPD1、2周组(P<0.01,0.05)。结论不同时期COPD大鼠肺组织中VEGF mRNA表达及肺血管的超微结构改变不同,提示VEGF可能参与了COPD血管内皮细胞损伤。Objectives To observe the dynamic changes of the expression of vascular endothelial growth factor(VEGF) mRNA in lung tissues and the uhrastructure of pulmonary vessels of rats in different periods of chronic obstructive pulmonary disease (COPD), and to explore the correlation between them. Methods Forty-eight healthy male rats were randomly divided into control group (n = 24 ) and COPD group (n = 24 ). The rats in COPD group were divided into COPD 1 week group, COPD 2 weeks group and COPD 4 weeks group, eight rats in each group. The models of the COPD were established by the improved passive smoking and lipopolysaecharide dropped into the trachea. The rats were killed at the first ,second and fourth week ,then the content of VEGF mRNA in lung tissues and the ultrastructure of pulmonary vessels of rats in different periods of COPD. Results With the progress of COPD, the uhrastructure of pulmonary vessels changed obviously with a trend of increasing gradually, and the expression of VEGF mRNA also increased gradually, which in COPD 1,2 and 4 weeks group were significantly higher than those in control group( P 〈 0.05,0.01 ) ;the expression of VEGF mRNA in COPD 2 weeks group was significantly higher than that in COPD 1 week group( P 〈 0.05 ) ;the expression of VEGF mRNA in COPD 4 weeks group were significantly higher than that in COPD 1,2 weeks group ( P 〈 0.05,0.01 ). Conclusion The expression of VEGF mRNA in lung tissues and the changes of uhrastructure of pulmonary vessels in different periods of COPD are different in rats, which shows that VEGF may be involved in the vascular endothelial cell injury during the course of COPD.
