摘  要:目的:观察洛伐他汀对大鼠心脏成纤维细胞多聚ADP核糖合成酶(PARP)-1活性及表达的影响。方法:采用PARP活性检测试剂盒和Western blot法检测洛伐他汀作用前后细胞内PARP-1的活性和蛋白表达水平。结果:使用洛伐他汀后,PARP-1的活性和蛋白表达均下调;再加用甲羟戊酸后,PARP-1的活性和表达下调被逆转。结论:洛伐他汀不仅可抑制血管紧张素Ⅱ刺激的心脏成纤维细胞PARP-1的活性及表达,还可在基础状态下抑制PARP-1的活性及表达,以上抑制作用都可以被甲羟戊酸逆转。Objective:To explore the effect of lovastatin treatment on activity and expression of poly (ADP- ribose) polymerase (PARP)-1 in cultured cardiac fibroblast. Method:The activity and expression of PARP-1 were measured by universal colorimetric PARP assay kit and Western blot assay. Result: The activity and expression of PARP-1 were down-regulated by lovastatin treatment. And this response could be reversed by addition of mevalonate. Conclusion:Lovastatin can obviously inhibit the activity and expression of PARP-1 induced by Ang II and under the normal condition. And these suppressive effects can be reversed by mevalonate.
