野黄芩苷对人舌鳞癌细胞的增殖抑制和Bcl-2蛋白表达的影响

摘　　要：目的:探讨野黄芩苷对人舌鳞癌细胞的增殖抑制作用及Bcl-2蛋白表达的影响。方法:用不同浓度的野黄芩苷处理体外培养的人舌鳞癌Tca8113细胞,用MTT法检测细胞增殖情况,Western-blot法检测Bcl-2蛋白表达情况。结果:野黄芩苷对Tca8113细胞的增殖有明显抑制作用,且在一定的时间和浓度范围内呈时间、浓度依赖性。Westerm-blot结果显示Bcl-2蛋白表达水平随野黄芩苷浓度的增大而逐渐下降。结论:野黄芩苷对人舌鳞癌细胞增殖具有明显的抑制作用,下调Bcl-2蛋白表达可能是野黄芩苷治癌的机制之一。Objective： To study the effects of scutellarin on the proliferation and Bcl-2 expression of human tongue carcinoma Tca8113 cells. Methods： Tca8113 cells were treated by scutellarin at 0（the control） ,80,120 and 160 μg/ml for 12, 24 and 48 h re- spectively. The cell proliferation was examined by MTF assay, Bcl-2 expression was tested by western bolt. Results： Scutellarin inhibi- ted the proliferation of Tca8113 ceils in a time-and dose-dependent manner. Western-Blot analysis showed that Bcl-2 protein expres- sion decreased progressively with the increase of scutellarin concentration. Conclusion： Scutellarin may inhibit the proliferation of Tca8113 ceils, and the mechanism may be related to down-regulation of Bcl-2 protein expression.
