年龄相关性白内障晶状体中PON1在糖尿病与非糖尿病患者的差异

摘　　要：目的糖尿病患者的年龄相关性白内障发病年龄轻、进展迅速。测定观察2型糖尿病患者及非糖尿病患者年龄相关性白内障品状体中对氧磷酶1（PONl）的表达情况。方法临床病例对照研究。对2010年3月至2011年3月在无锡人民医院眼科就诊的36例患有2型糖尿病的年龄相关性闩内障患者组成实验组，50例非糖尿病年龄相关性白内障患者组成对照组，行白内障手术取出晶状体，利用ELI—SA法检测2型糖尿病患者与非糖尿病者的年龄相关性白内障中PONl的表达量，以对氧磷为底物采用酶活性法检测PONl的＇活ll生。结果2型糖尿病患者较非糖尿病者年龄相关性白内障品状体中PONl活性偏低差异有统计学意义（t=4．38，P〈0．01），而两组PONl酶的量差异无统计学意义（t=0．2305，P=0．92）。结论PONl酶量在2型糖尿病患者和非糖尿病患者年龄相关性白内障品状体中无差异，但其酶活性存在差异，推测可能的原因是糖尿病改变了PONl酶的空间结构，有待进一步深入的研究。Objective To observe the expression of paraoxonase-1 （PONI） between type 2 diabet- ic and normal age-related cataract. Methods The lenses were obtained from 36 cases of type 2 dia- betic patients and 50 age-related cataract patients during the surgery. The ELISA was used to detect expression of PON1 and enzyme activity assay was selected to analysis activity of PON1. Results The enzymatic activity of PON1 in type 2 diabetic age-related cataract was lower than normal age-re- lated cataract （t =4.38, P 〈0.01）, however, the amount of PON1 in two groups had no difference （t =0.2305, P =0.82）. Conclusions Lower enzymatic activity of PON1 is detected in type 2 diabet- ic age-related cataract, which may be explained for the changed structure. In-depth study should be carried out in the future.
