左旋多巴诱导PC12细胞凋亡实验在治疗帕金森病效果减退(剂末现象)机制探讨中的意义

摘　　要：目的通过进行左旋多巴诱导PC12细胞凋亡实验探讨左旋多巴治疗帕金森病效果减退(剂末现象)的机制。方法选取大鼠肾上腺嗜铬细胞瘤(PC12)细胞为多巴胺(DA)神经元细胞模型,给予左旋多巴处理,应用流式细胞术以及DNA凝胶电泳图像分析技术观察左旋多巴对PC12细胞凋亡的影响。结果 DNA琼脂糖凝胶电泳结果显示,对照组、50μmol/L组、100μmol/L组、150μmol/L组和200μmol/L组细胞DNA片段化比例分别为(2.55±0.24)%、(7.49±0.41)%、(13.41±1.62)%、(25.26±3.29)%和(40.01±5.18)%,方差分析检验,5组间比较差异有统计学意义(F=167.67,P﹤0.05),进一步经q检验(Newman-Keals法),5组间分别两两比较差异有统计学意义(P﹤0.05)。流式细胞术检测结果显示,对照组、50μmol/L组、100μmol/L组、150μmol/L组和200μmol/L组细胞凋亡率分别为(2.31±0.25)%、(8.14±0.41)%、(12.39±0.47)%、(24.29±0.38)%和(39.12±1.12)%,方差分析检验,5组间比较差异有统计学意义(F=3476.49,P﹤0.05),进一步经q检验(Newman-Keals法),5组间分别两两比较差异有统计学意义(P﹤0.05)。结论左旋多巴可诱导PC12细胞凋亡,该机制可能参与左旋多巴治疗帕金森病剂末现象的病理过程。OBJECTIVE To discuss the mechanism of L-dopa treatment for Parkinson＇s disease effects fade （wearing-off） by the L-dopa induced PC12 cell apoptosis experiment. METHODS The rat adrenal pheochromocytoma （PC12） cells were select- ed as dopamine （DA） neurons model and were givenL-dopa treatment. The effect L-dopa for PC12 cell apoptosis was observed by the application of flow cytometry and DNA gel electrophoresis image analysis technology. RESULTS The DNA agarose gel electrophoresis results show that, the DNA fragmentation ratio of the control group, 50 ~ moL/L group, 100 ~ mol/L group, 150 Iz mol/L group and 200 ~ mot/L group cell were （2.55 ＋ 0.24） %, （7.49 ＋ 0.41） %, （13.41 ＋ 1.62） %, （25.26 ＋ 3.29） % and （40.01 ~ 5.18） %, by the analysis of variance test. The differences among five groups were statistically signifi- cant （F = 167.67, P 〈 0.05）. Additionally, the respectively pairwise comparison in the five groups were all statistically signifi- cant （P 〈 0.05） by using the q test （Newman - Keals method）. Flow cytometry deteetion＇s result shows, the apoptotic rate of the control group, 50 μ mol/L group, 100 μ mol/L group, 150 μ mol/L group and 200 μ mol/L group cell were （2.31 ± 0.25） %, （8.14± 0.41） %, （12.39± 0.47） %, （24.29 ± 0.38） % and （39.12 ± 1.12） % by the analysis of variance test. The differences among the five groups were statistically significant （F = 3476.49, P 〈 0.05）. By the q test （ Newman - Keals method）, the respectively pairwise comparisons in the five groups were all statistically significant （P 〈 0.05）. CON- CLUSION The L-dopa can induce PC12 cell apoptosis. This mechanism may be involved inpathological process of the wearing- off of L-dopa treatment for Parkinson＇ s disease.
