摘  要:目的探讨去甲基化药物5-氮-2-脱氧胞苷对体外培养表皮癌细胞株A431细胞Gadd45α表达及细胞凋亡的影响。方法体外培养A431细胞,采用10μmol/L 5-氮-2-脱氧胞苷处理,药物处理前及处理48h后分别采用RT-PCR、Western印记法及免疫细胞化学法检测A431细胞Gadd45αmRNA和蛋白的表达情况,采用流式细胞仪检测药物处理前、后A431细胞凋亡率。结果药物处理48h后,A431细胞Gadd45αmRNA及蛋白表达均高于处理前(P<0.05);A431细胞早期凋亡率、晚期凋亡率及总凋亡率高于处理前(P<0.05)。结论 DNA甲基化参与调控A431细胞Gadd45α基因的表达,5-氮-2-脱氧胞苷具有诱导A431细胞凋亡的作用。Objective To study the effect of 5-aza 2'-deoxycytidine (5-Aza-dC) on the expression of Gadd45a and apoptosis of A431 cells. Methods Reverse transcription PCR, Western blot and immunocytochemistry were used to detect Gadd45a mRNA and protein expressions in A431 cells after treatment with 10 μmol/L 5-aza-dC for 48 hours, and flow cytometry was used to measure the cell apoptosis rate of A431 ceils. Results After treatment with 10 μmol/L 5 aza-dC for 48 hours, Gadd45a expression of A431 cells at both mRNA and protein levels, the early apoptosis rate, late apoptosis rate and total apoptosis rate were significantly higher than those in untreated group (P〈 0.05). Conclusion DNA methylation may be involved in the modulation of aberrant Gadd45a gene expression in A431 cells, and 5-aza-dC could induce the apoptosis of A431 cells.
