摘  要:目的初步探讨硫化氢(H2S)对内毒素(LPS)所致急性肺损伤(ALI)大鼠PMN功能的影响机制。方法将112只大鼠,随机分为4组每组28只,即盐水对照组(A组)、LPS组(B组)、LPS+硫氢化钠(NaHS)组(C组)、LPS+炔丙基甘氨酸(PPG)组(D组),再根据给药时间分成两亚组(4h组和8h组)。均于给药4h(14只)或8h(14只)后经颈总动脉放血处死动物,留取血标本应用流式细胞学方法检测血PMNCDllb;在每一亚组各取7只大鼠留取肺组织,应用邻连茴香胺法测定肺组织髓过氧化物酶(MP0)活性、双抗体夹心ABC-ELlSA法检测肺组织中黏附分子-1(ICAM—1)的含量表达的变化,光镜下观察肺组织形态学变化并计算肺泡损伤数比值(IQA)作为肺损伤的组织学定量评价指标;剩余大鼠行支气管肺泡灌洗(BAL)并在光镜下计量支气管肺泡灌洗液(BALF)中PMN数目。结果气管内滴注LPS后可引起大鼠肺损伤、IQA升高、BALF中PMN的数目增多、MPO活性及ICAM-1蛋白表达上调,预先给予NaHS可显著减轻肺损伤、IQA下降、BALF中PMN的数目减少、MPO活性及ICAM-1蛋白表达下降,预先给予PPG可加重肺损伤、IOA升高、BALF中PMN的数目更加增多、MPO活性及ICAM-1蛋白表达上调(P均〈0.0l或P均〈O.05,F值=22.256或35.168;F值-32.760或54.722;F值=40.133或45.263;F值-85.223或98.187);血PMNCDllb与BALF中PMN计数存在显著正相关(r-0.789,P均〈O.01),有统计学意义。结论H2S可通过减少ICAM-1,CDIib的表达、降低MPO活性来抑制ALI大鼠PMN的功能。Objective To observe the effect of hydrogen sulfide (H2S) on polymorpbonuclear (PMN) function in lipopolysac- charide (LPS)-induced acute lung injury (ALI) rats. Methods 112 Sprague-Dawley rats were randomly divided into four groups: sham-operated group,model group (LPS were instilled intratracheally to induce ALI), LPS+ Naris (H2S donor) group,and LPS+PPG (propargylglycin,CSE inhibitor) group. Then according to the dosage time divided into two sub- groups (group 4h and group 8h). All the rats were observed at 4 h (14 rats) or 8 h (14 rats)after agent administration. The blood PMN CDtlb expression was detected by Flow cytometry. Seven rats of each subgroups,Lung tissue myeloperoxidase (MPO) activity was used to detect by Neighbors even fennel amine method. ICAM-1 protein content in lung tissues was cal- culated to measure by double antibody sandwich elisa method. Lung histomorphological changes were observed under micro- scope and index of quantitative assessment (IQA) was calculated to measure the lung injury severity. Seven rats of each sub- groups were received bronchoalveolar lavage. PMN number in Bronchoalveolar lavage fluid (BALF) was counted under light microscope. Results Compared with sham-operated group, severe injuries of lung tissues and a raised IQA, a raised PMN number in BALF, expression of ICAM-1 protein and expression of MPO protein were observed in rats treated with LPS. Pre- administration of NariS not only alleviated the injuries induced by LPS but also decreased IQA,PMN number in BALF, ex- pression of ICAM-1 protein and expression of MPO protein. Pre-administration of PPG exacerbated the injuries induced by LPS. Also,increased IQA,PMN number in BALF, expression of ICAM-1 protein and expression of MPO protein were ob- served.
