摘 要:目的研究提高菌种的反硝化能力及提高污水氮去除率的方法,确定菌株的最佳生长条件,选育出纯种好氧反硝化菌株.方法采用平板稀释法和NO3--BTB培养基从污泥中分离纯化好氧反硝化菌株,以紫外线为诱变剂对菌种进行诱变,并复筛得到反硝化能力增强的菌株作为优势菌.设计四因素三水平正交试验,四因素为pH值、温度、碳源、ρ(C)/ρ(N),通过正交试验确定正突变优势菌株的最佳生长条件.结果紫外诱变选育得到反硝化能力增强的好氧反硝化菌株,总氮去除率提高到85.74%,亚硝酸盐积累明显减少,菌株最佳生长条件pH值为7、温度为25℃、ρ(C)/ρ(N)为10.结论紫外诱变育种能提高好氧反硝化菌种的反硝化能力,温度和ρ(C)/ρ(N)对菌株生长影响最大.An aerobic denitrification bacteria was isolated from the sewage sludge and mutagenized by UV-ir- radiation to improve the capability of aerobic denitrification and removal efficiency of nitrogen. Meanwhile, the optimal culture conditions and aerobic denitrification performance of the strain were investigated. Hate dilution method and NO3-BTB selective medium were used to screen. Then the UV lamp irradiated bacteria suspension of aerobic denitrification bacteria to mutagenize. And a high nitrogen-removal mutant was ob- tained. Orthogonal experiment of four factors and three levels was designed to ascertain the optimal culture condition in the high nitrogen-removal mutant. The four factors were pH value, temperature, carbon-nitrogen ratio and carbon source. Removal rate of total nitrogen of the high nitrogen-removal mutant is 85.74% and the accumulation of nitrite reduced significantly. The optimal culture conditions for maximum denitrification were:pH 7,25 ℃ ,p(C)/p(N)ratio = 10, and carbon source was glucose. The aerobic denitrification per- formance of the high nitrogen-removal mutant was mostly influenced by temperature.
