摘 要:目的比较小凹蛋白、生长抑素、姜黄素单独及联合应用抗肝纤维化的作用并探讨其作用机制。方法将3种药物分别单独作用于大鼠肝纤维化模型和大鼠肝星状细胞。再将小凹蛋白分别联合生长抑素及姜黄素作用于大鼠肝纤维化模型和大鼠肝星状细胞。检测血透明质酸酶(HA)、Ⅳ型胶原含量(CⅣ);MTT法检测肝星状细胞增殖抑制率;免疫组化法检测转化生长因子-β1(TGF-β1)的表达。结果生长抑素及姜黄素均有抗肝纤维化作用,小凹蛋白可增强姜黄素抗肝纤维化的作用(P<0.05),但未观察到其增强生长抑素抗肝纤维化的作用(P>0.05)。小凹蛋白与姜黄素均可抑制TGF-β1表达(P<0.05),两者联用,抑制作用更强(P<0.05)。结论抑制TGF-β1可能是姜黄素与小凹蛋白抗肝纤维化的共同作用途径之一。Objective To compare the anti-hepatic fibrosis effects of caveolin-1,somatostatin and curcumin,and explore the possible mechanism.Methods Caveolin-1,somatostatin and curcumin were given to rats with hepatic fibrosis and added to the culture of hepatic stellate cells(HSC)respectively alone and by combination.Transforming growth factor beta 1(TGF-β1) expression was detected by immunohistochemistry.HSC proliferation was evaluated by MTT assay.The levels of blood hyaluronic acid(HA) and type Ⅳ collagen(CⅣ) were also analyzed.Results Somatostatin and curcumin had anti-hepatic fibrosis effects(P0.05).Caveolin-1 could enhance the anti-hepatic fibrosis effect of curcumin(P0.05),but could not enhance the effect of somatostatin(P0.05).Caveolin-1 or curcumin alone could inhibit the expression of TGF-β1(P0.05);when combined the two,stronger inhibition effect could be observed(P0.05).Conclusion Inhibition of TGF-β1 might be a part of the common path of caveolin-1 and curcumin to inhibit hepatic fibrosis.
