摘 要:目的研究肾移植术后患者ABCB1基因多态性对环孢素A(cyclosporin A,CsA)所致肝功能异常的影响。方法采用聚合酶链反应(polymerase chain reaction,PCR)-限制性片段长度多态性(restriction fragment lengthpolymor-phism,RFLP)方法(PCR-RFLP),对339名服用CsA的肾移植患者进行ABCB1基因1236C>T、2677G>T/A和3435C>T多态性检测,根据肾移植患者发生肝脏损伤的情况分为CsA致肝功异常组、其他原因肝损伤组和对照组。结果在339名肾移植患者中,ABCB1基因1236C>T、2677G>T/A和3435C>T突变等位基因频率分别为64.25%、51.54%和35.75%。ABCB1基因1236C>T、2677G>T/A和3435C>T多态性在CsA致肝功异常组、其他原因肝损伤组和对照组3组之间的分布频率无统计学差异(P>0.05)。结论 ABCB1基因1236C>T、2677G>T/A和3435C>T多态性与CsA所致的肝功异常无明显相关性。Objective To investigate the effects of At〉B1 genetic polymorphism on cyclosporine (CsA)-induced liver injury in Chinese renal transplant recipients. Methods ABCB1 1236C〉T, 2677G〉T/A and 3435C〉T genotypes were determined by polymerase chain reaction (PCR) followed by restriction fragment length polymorphism (RFLP) analysis in a total of 339 renal transplant recipients receiving CsA. All the subjects were divided into CsA-induced liver in- jury group, non-CsA-induced liver injury group and the control group according to the liver injury occurrence. Results In this study of 339 renal transplant recipients, the frequencies of ABCB1 1236C〉T, 2677G〉T/A and 3435C〉T mutation allele were 64. 25% ,51.54% and 35.75% respectively. The distribution frequencies of ABCB1 1236C〉T, 2677G〉T/A and 3435C〉T mutation allele had no significant difference among three groups. Conclusion There is no association be-tween ABCB1 genetic polymorphism and cyclosporine-induced liver injury in Chinese renal transplant recipients.
