摘 要:目的:研究补气活血法对大鼠失神经肌萎缩的影响。方法:选雄性SD大鼠60只,随机分为6组:假手术组、模型组、阳性对照组、中药低、中、高剂量组。后5组采用胫前肌施夹伤手术,建立大鼠腓总神经夹伤模型。造模后,术后各组每日分别灌胃给药,持续18天。全部处死取材,观察各组大鼠左侧胫前肌形态学变化,并采用RT-PCR法观察大鼠左侧腓肠肌TGF-β1mRNA表达水平。结果:中药各组骨骼肌横切面的性状变为棱角形,肌核彼此密集,细胞膜破裂,但不明显,脱水现象有所减少,细胞有所破坏,但破坏程度不大;阳性对照组肌纤维崩解,细胞破裂明显,甚至出现自溶现象,细胞破坏程度较大,但其脱水现象减轻,棱角出现。各组大鼠胫前肌TGF-β1mRNART-PCR结果显示,与模型组相比,中药低、中、高剂量组胫前肌TGF-β1mRNA表达水平均降低(P<0.05,P<0.01);高剂量组胫前肌中TGF-β1mRNA转录水平与中药低剂量组比较,均有显著性(P<0.05);且中药高剂量组胫前肌TGF-β1mRNA表达与阳性对照组相比无显著差异(P>0.05)。结论:补气活血法能有效防治大鼠失神经肌萎缩,其作用机制可能与其干预胫前肌中TGF-β1mRNA表达有关。Objective : Objective : To study the effect of method of invigorating qi to promote blood flow on rat' s denervated muscle atrophy. Methods: 60 male SD rats were randomly divided into 6 groups, which were sham operation group, model group, positive control group, low, middle and high dose of treatment group. The models of common-peroneal-nerve clamp-injury were made by the operation of tibialis-anterior-muscle clamp-injury. Then they had been given to intragastric administration for 18 days. At last they were executed, and the morphology of their left tibial muscles was observed in each group, and the expression of TGF-β1mRNA in right gastrocnemius muscle was detected with RT-PCR. Result: In each treatment group the cross section of the skeletal muscle be- came into corniform, the cellular nuclei of the muscle ceils were intensive, the phenomenon of cell membrane rup- ture showed up but not obviously, dehydration was decreased, and the cells were damaged but few. In positive control group the muscle fiber disaggregated, the plasmatorrhexis was obvious and even emerged autolysis, and the cells damaged greatly. But its dehydration was lessened. TGF-β1 mRNA expression of the tibialis anterior muscle in low, middle and high dose of treatment group decreased more than in model group (P 〈 0. 05, P 〈 0. 01 ). Compared with low dose of treatment group the TGF-β1 mRNA transcriptional level was significant in high dose of treatment group (P 〈 0. 05 ). And there was not difference between high dose of treatment group and positive con- trol group on the expression of TGF-β1mRNA (P 〉 0. 05 ). Conclusion: Method of invigorating qi to promote blood flow can prevent rat from denervated muscle atrophy effectively, and the possible mechanism of action may be re- lated to its expression of TGF-β1 mRNA in tibialis anterior muscle.
