黄芪提取物抑制胃癌细胞对腹膜间皮细胞损伤的实验研究

摘　　要：目的观察黄芪提取物抑制胃癌细胞上清对腹膜间皮细胞的凋亡作用。方法将胃癌细胞(MKN45)上清单独或与黄芪提取物联合作用于人腹膜间皮细胞HMrSV。光镜下观察人腹膜间皮细胞形态学变化;MTT检测黄芪作用后人腹膜间皮细胞生长曲线;流式细胞仪检测细胞凋亡率,荧光显微镜观察细胞凋亡情况。结果 MTT及流式细胞仪检测出胃癌细胞(MKN45)上清诱导人腹膜间皮细胞凋亡呈时间依赖性;荧光显微镜观察胃癌细胞(MKN45)上清作用后,间皮细胞出现典型凋亡特征。结论黄芪提取物明显抑制胃癌上清促间皮细胞凋亡作用。Objective To observe the effect of astragalus extract on inhibition of the peritoneal mesothelial cells apoptosis by gastric cancer cells. Methods By using the supernatant of MKN45 alone or with astragalus extract to affect HMrSV, the morphological changes of human peritoneal mesothelial cells was observed by light microscope, astragalus in human peritoneal mesothelial cells growth curve was detected by M＇IW, cell apoptosis was observed by flow cytometry and fluorescence microscope. Results The MTT and flow cytometry showed that supernatant of MKN45 can induce apoptosis of human peritoneal mesothelial ceils in a time-dependent manner. The typical apoptotic features was found by fluorescence microscope. Conclusion Astragalus extract significantly inhibits gastric carcinoma supernatant of mesothelial cells apoptosis.
