摘 要:目的探讨大鼠髁突软骨发育过程中IGF-1、IGF-1R及凋亡相关因子Bcl-2、Bax的表达、分布以及可能发挥的作用。方法获取出生后1、7、14、28d SD大鼠髁突,甲苯胺蓝染色观察软骨结构。免疫组化染色检测IGF-1、IGF-1R、Bcl-2、Bax在各年龄组SD大鼠髁突软骨的分布。Western blot及real-time PCR分别检测各年龄组软骨细胞内IGF-1、IGF-1R、Bcl-2、Bax蛋白及mRNA的表达。结果甲苯胺蓝染色结果显示随年龄递增,大鼠髁突软骨逐渐变薄,宽度增加。免疫组化染色显示IGF-1、IGF-1R、Bcl-2在增殖层至肥大软骨细胞浅层分布最明显,Bax主要表达在成熟层及肥大层。Real time PCR及Western blot结果表明出生后的IGF-1 mRNA的表达逐渐降低,至28d时表达开始增强,IGF-1R蛋白及mRNA出生后稍有波动但表达相对平稳,Bcl-2、Bax蛋白及mRNA表达逐渐增强,至出生14d时表达开始下降(P<0.05)。结论 IGF-1、IGF-1R及凋亡相关因子Bcl-2、Bax的表达及分布相关,共同参与调控髁突软骨的发育。Objective To explore the expression and distribution of IGF-1、IGF-1R and apoptosis-associated factors Bcl-2、Bax and their possible functions during the development of rat condyalr cartilage.Methods The 1、7、14 and 28-day-old rat condyles were obtained and their shape and structures were observed with toluidine blue stain method.The distribution of IGF-1、IGF-IR、Bcl-2 and Bax proteins on 1-、7-、14-and 28 day-old condylar cartilage were evaluated by immunohistochemistry.The expression of IGF-1、IGF-1R、Bcl-2、Bax mRNA and proteins in each group were investigated by real time PCR and western blot analysis.Results Toluidine blue staining showed the rapid developmental changes of rat condylar cartilages about thinner cartilage and thicker subchondral bone,and they were followed by rapid growth in condylar size.The immunohistochemical staining was positive for IGF-1、IGF-1R、Bcl-2,which was mainly located from the proliferation to upper hypertrophic layers.The distribution of Bax proteins were mainly on mature chondrocyes layers and hypertrophic layers.The result of real time PCR and western blot analysis showed that IGF-I mRNA expression levels strated to decrease immediately after the animals' birth,and increased from 28 days.IGF-IR protein and mRNA expression levels were relatively well maintained although there was some variation.Bcl-2、Bax protein and mRNA expression levels reached a peak at 14 days and thereafter decreased(P0.05).Conclusion IGF-1、IGF-1R and apoptosis-associated factors Bcl-2、Bax show analogously temporal expression and distribution,play a regulatory role in condylar cartilage development.
