摘 要:目的在中医理论指导下建立SD大鼠寒积里实证动物模型并对其进行评价。方法应用先冰水后高岭土灌胃方法制作寒积里实证动物模型。观察模型动物的一般生物学特征变化,进行模型动物的排便和尾部温度测量实验,检测模型动物的肠神经递质、胃肠激素、cAMP和cGMP等客观指标。结果造模后,模型动物的一般生物学特征表现为:披毛蓬松、好聚集饲养笼一端、大便干燥,摄食量降低;与空白对照组比较,模型组动物的6 h排便粒数明显减少,尾部温度明显降低,胃动素、血管活性肠肽、P物质分泌紊乱,cAMP/cGMP降低,经大黄附子汤治疗后,上述症状和指标改善。结论应用先冰水后高岭土灌胃方法能复制出稳定的SD大鼠寒积里实证模型。Aim To establish and evaluate Sprague-Dawley(SD) rats model with cold accumulation and internal excess under the guidance of traditional chinese medicine theories.Methods Rats model with cold accumulation and internal excess was induced by oral administration of ice water firstly,then kaolin.Changes of the biological characteristics of the model animals,the excretion,tail temperature measurement and the detection of intestinal neurotransmitters,gastrointestinal hormone,cAMP,cGMP and other objective indicators were observed.Results The general biological characteristics of the model animal performance were performed as: puffed and withered hair,gathering at one end in cages,dry stools and reduced food consumption.Compared with the control group,defecation grains of model group reduced significantly within six hours.Meanwhile tail temperature reduced significantly,and motilin,vasoactive intestinal peptide,substance P secretion were in disorder and cAMP/cGMP reduced.After being treated with Dahuang Fuzi Tang,symptoms and indicators were improved markedly.Conclusion SD rats model with cold accumulation and internal excess can be stably established by administration of ice water firstly,then kaolin.
