摘 要:目的:通过研究结缔组织生长因子(CTGF)基因对人乳腺癌MCF-7细胞株基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶组织抑制因子-2(TIMP-2)表达的影响,深入探讨CTGF在乳腺癌侵袭和转移中的作用机制。方法:采用脂质体介导法,将CTGF基因瞬时转染体外培养的人MCF-7细胞,并用免疫荧光、RT-PCR和蛋白质印迹法检测其转染成功与否;再采用蛋白质印迹法检测转基因MCF-7 MMP-2及TIMP-2表达改变。结果:与对照组相比,转染CTGF基因的MCF-7,其CTGF mRNA和蛋白表达均显著增高,MMP-2蛋白表达水平显著增高,而TIMP-2蛋白表达水平显著降低。结论:CTGF可能通过增加乳腺癌细胞MMP-2表达及抑制TIMP-2的生成而起到促进乳腺癌侵袭和转移的作用。Objective: To investigate the effect of connective tissue growth factor (CTGF) on the expres- sion of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in human breast cancer line MCF-7. This study was designed to explore the underlying mechanism for the role of CT- GF in the development of breast cancer. Methods: Lipofectin method was used to transfect CTGF vectors into MCF-7; immunofluorescence, RT-PCR and Western blot analysis were used to detect their transfeetion efficiency. The expressions of MMP-2 and TIMP-2 were determined by Western blot analysis. Results: Im- munofluorescence, RT-PCR and Western blot analysis showed a successful transfection of CTGF vectors into MCF-7. MCF-7 transfected with CTGF gene showed an increased expression of MMP-2 protein significantly, compared with control groups. On the other hand, MCF-7 transfected with CTGF gene showed a decreased expression of TIMP-2 protein significantly, compared with control groups. Conclusion: It is possible that CTGF could promote the development of breast cancer by effectively enhancing the expressions of MMP-2 and TIMP-2 in MCF-7 cells.
