仁术健胃颗粒干预脾气虚证CAG大鼠对COX-2和EGFR表达的实验研究

摘　　要：目的:通过观察仁术健胃颗粒对慢性萎缩性胃炎(CAG)脾气虚证大鼠COX-2、EGFR的影响,探讨仁术健胃颗粒预防和治疗慢性萎缩性胃炎的机制。方法:采用N-甲基-N-硝基-N-亚硝基胍(MNNG)和40%乙醇灌胃建立大鼠慢性萎缩性胃炎(CAG)模型,同时以破气苦降,饥饱失常和疲劳训练进一步建立CAG脾气虚证大鼠模型。将CAG脾气虚证大鼠随机分为模型组、胃复春组和仁术健胃颗粒3个剂量组。给予相应的药物连续治疗5周,正常组给予等量NS。实验末检测各组大鼠胃组织中COX-2和EGFR的表达。结果:CAG脾气虚模型组的COX-2在胃腺体细胞胞浆中表达明显,EGFR从基底部到胃黏膜表层表达明显且范围广,与正常组比较有显著性差异(P<0.01)。采用仁术健胃颗粒干预后COX-2,和EGFR表达明显减弱(P<0.01;P<0.05)。结论:仁术健胃颗粒治疗CAG脾气虚证与降低胃组织中COX-2和EGFR的表达,这可能是其治疗CAG脾气虚证的重要机制之一。Objective： To observe the effect of Renzhu Jianwei Granule expressions of COX -2, MMP -2 and EGFR of chronic atrophic gastritis （CAG） rats with spleen - qi deficiency to explore the mechanism of prevention and treatment of chronic atrophic gastritis. Methods ：N - methyl - N - nitro - N - nitrosoguanidine （MNNG） and 40% ethanol garage were used to establish the chronic atrophic gastritis （CAG） model, and at the same time, reliveing Qi stagnation and de- scending with bitter drugs as well as irregular eating and fatigue training were used to establish model of spleen - qi defi- ciency. The CAG spleen - qi deficiency rats were randomly divided into model group, Weifuchun group and Renzhu Jian- wei granule three dose groups. The corresponding drug treatment were given for five consecutive weeks, normal control group were given NS. At the end of the experiment, expressions of COX - 2 and EGFR in gastric tissue of rats in each group were observed and compared. Results ： The COX - 2 expression in the cytoplasm of gastric body of CAG of spleen - qi deficiency model group was obvious;EGFR expression had a wide range fi＇om the basal to the gastric mucosal surface, there was significant difference compared with the normal group （ P 〈 0.01 ）. After treated with Renzhu Jianwei granule, COX - 2 and EGFR expressions were significantly decreased （ P 〈 0. 01 ; P 〈 0.05 ）. Conclusion ： Renzhu Jianwei granule can CAG of spleen - qi deficiency, and decrease the expressions of COX - 2 and EGFR in gastric tissue, which may be the mechanism of treatment of CAG of spleen - qi deficiency syndrome.
