摘  要:目的探讨辐射诱发的人鼻咽低分化鳞癌细胞株(CNE-2)凋亡率和Bak、Bid、Bcl-w、Bcl-xl转录水平的影响。方法体外培养CNE-2,应用流式细胞术及RT-PCR方法检测不同辐射剂量(0、2、4、6、8Gy)下CNE-2凋亡率及凋亡相关基因Bak、Bid、Bcl-w、Bcl-xl的转录水平。结果 CNE-2照射后Bak表达上调,与辐射剂量及晚期凋亡率呈正相关;Bcl-w表达下调,与辐射剂量及晚期凋亡率呈负相关;Bcl-xl表达下调,与辐射剂量及晚期凋亡率无相关性;Bid在不同剂量点转录水平差异均无统计学意义。结论 Bak、Bcl-w、Bcl-xl可能参与了辐射诱发的CNE-2细胞凋亡的调控。Objective To investigate the relationship between the mRNA expression of Bak, Bid, Bcl-w, Bcl-xl and radiation induced apoptosis in poor differentiated nasopharyngeal carcinoma (NPC) cell line CNE-2. Methods The NPC cell line CNE2 were in vitro culture, cellular apoptosis was measured by flow cytometry, reverse transcription polymerase chain reaction (RT-PCR) was used to detect mRNA expres sions of Bak, Bid, Bcl-w and Bcl-xl respectively before and after radiation (the dosage were 2, 4, 6, 8 Gy, respectively) with linear accelerator. Results In CNE 2, the mRNA e positively correlated with the dose of radiation and late apoptosis rate , xp th ression of Bak was increased and e mRNA expression of Bcl-w was decreased and negatively correlated with the dose of radiation and late apoptosis rate, the mRNA expression of Bcl-xl was decreased and unrelated with the dose of radiation and late apoptosis rate, the mRNA expression of Bid in different dose group was not statistically significant. Conclusion Bak, Bcl-w Bcl-xl may participate in the regulation of radiation induced apoptosis in CNE-2.
