摘 要:目的:观察Smac基因过表达对胃癌细胞株化疗敏感性的影响。方法:采用脂质体lipofec-tamine2000介导的方法,将Smac基因转入胃癌细胞SGC-7901,RT-PCR和Western-blot法检测未转染对照组、空载体pcDNA3.1转染对照组、pcDNA3.1-Smac转染组胃癌细胞中Smac基因的表达。选用紫杉醇(1、5、10μg/mL)处理转染前后的胃癌细胞,四甲基偶氮唑蓝(MTT)比色法检测细胞增殖活性,倒置显微镜下观察细胞形态变化并摄片。结果:同未转染对照组和空载体pcDNA3.1转染对照组比较,pcDNA3.1-Smac转染组SGC-7901细胞Smac mRNA和蛋白表达水平显著增高(P<0.01);紫杉醇处理24、48 h后,pcDNA3.1-Smac转染组细胞生长抑制率增加(P<0.01);使用化疗药物后,细胞明显变圆、折光增强、漂浮细胞增多,pcDNA3.1-Smac转染组细胞形态变化最为显著。结论:转染外源性Smac基因并使其在胃癌细胞中过表达,能提高SGC-7901细胞对化疗药物的敏感性。Objective: To explore the effect of overexpressed Smac gene on chemotherapeutic sensitivity of gastric cancer cell line. Method: By lipofectamine2000, Smac gene was transfectedinto gastric cancer cell line SGC-7901. Cellular Smac gene expression were determined by reverse transcription-polymerase chain reaction(RT-PCR) and Western blot analysis. Result: RT-PCR and Western blot results demonstrated smac mRNA and protein levels of SGC-7901/Smac cells were significantly higher than those of SGC-7901 and SGC-7901/neo (P 〈 0.01). Conclusion: Trans- fection of extrinsic Smac gene results in its over-expression in SGC-7901,which could increase the chemotherapeutic sensitivity of SGC-7901 cell.
