摘 要:Ras蛋白是调节细胞生长和增殖信号通路的重要元件。当Ras变异时,将信号转导到下游信号元件可能引起细胞的异常增殖,导致肿瘤发生。抑制Ras及其下游信号可抑制细胞增殖和促进肿瘤细胞凋亡,研究表明Ras信号通路在食管肿瘤的发病过程中起着重要作用。Ras is best known for the ability of regulating cell growth, proliferation and differentiation. Muta- tions in Ras are associated with abnormal cell proliferation, resulting in the incidence of all human cancers. Mitogen-activa- ted protein kinase (MAPK) signaling pathways are the most well described pathways in carcinogenesis, which has been i- dentified as a key downstream effector in Ras signaling as well as playing important roles in prognosis of tumors. Recently, evidence has gradually accumulated to demonstrate that mutation in Ras or aberrant expression of MAPK has profound effects on the incidence of esophageal carcinoma, and applications of some chemotherapeutic drugs can not lead to the ex- pectant function. Further understanding of the relevant molecular mechanisms of Ras-MAPK signaling pathways will be helpful for development of efficient targeting therapeutic approaches to the treatment of esophageal cancer. In this article, the advances of Ras-MAPK signaling pathways in esophageal carcinoma are reviewed.
