摘 要:目的探讨大鼠在生理性衰老过程中肠道敏感性、血清及脑肠轴Ghrelin水平的变化及其意义。方法将健康SD大鼠按3月龄、9月龄、18月龄和24月龄分为4组,每组各6只。采用直肠球囊扩张引起大鼠腹肌收缩产生的曲线下面积评价肠道敏感性;采用双抗体夹心酶联免疫方法检测大鼠血清及脑肠轴组织中的Ghrelin含量。结果①球囊直肠扩张后,3月龄组大鼠腹肌收缩产生的曲线下面积最小,而随着月龄的增大,曲线下面积逐渐增大,在中等容量及高容量扩张后更为显著(P均<0.05);②3月龄组大鼠血清Ghrelin含量最低,9月龄组最高,之后有逐渐降低的趋势,但各组间比较无统计学差异(P>0.05);大鼠全脑组织中Ghrelin含量3月龄组最低(P<0.01),之后增高,但9月龄、18月龄、24月龄组间比较差异无统计学差异(P>0.05);③胃肠道组织中Ghrelin含量随着大鼠月龄的增大而逐步增高(P均<0.05);同月龄组大鼠胃肠道组织胃体Ghrelin含量最高(P<0.01),空肠、回肠与胃体的含量相比减少(P<0.05),结肠较空肠、回肠降低(P<0.05),乙状结肠与直肠交界部Ghrelin含量最低(P<0.05)。结论在大鼠生理性衰老过程中,肠道敏感性随月龄的增加而增高;血清Ghrelin含量随月龄的增加而降低,脑肠轴组织中Ghrelin含量随月龄的增加有增高的趋势,但是变化不是很明显。这些变化导致的胃肠道感觉与运动异常可能参与了老年人群胃肠功能性疾病的发生和发展。Objective To observe changes of intestinal sensitivity and Ghrelin levels in serum and brain-gut axis in the physiological aging process of SD rats.Methods Twenty-four healthy SD rats were divided into four groups according to the age of three months,nine months,eighteen months and twenty-four months,respectively,with six rats in each group.Rectal saccule dilatation which can cause abdominal muscle contraction was applied.Intestinal sensitivity was estimated by the area under the curve(ANC) generated by muscle contraction.The contents of Ghrelin in serum and brain-gut axis were measured by ELISA.Results ① After rectal saccule dilatation,3-month-old rats showed the smallest ANC,and the ANC was expanded with the increase of age,especially in the medium and high intensity of dilatation(all P0.05).② Ghrelin level in serum was the lowest in the 3-month group and the highest in the 9-month group.After that,the serum level of Ghrelin tended to decrease,but without significant difference(P0.05).③ Ghrelin level in gastrointestinal tissues tended to increase gradually with age(all P0.05),and the level in the same age group was as follows: the highest in the body of the stomach(P0.01),followed by the jejunum and the ileum(P0.05),and the lowest in the sigmoid colon and the rectum junction(P0.05).Conclusion Intestinal sensitivity of SD rats is increased in the process of physiological aging;Ghrelin level in serum is decreased and that in brain-gut axis is increased in the physiological aging process on the whole.These changes may cause gastrointestinal sensory and motor abnormalities,and are possibly involved in the occurrence and development of functional gastrointestinal disorders in old people.
