摘 要:目的通过大鼠实验模型及人胃黏膜细胞培养技术探讨AMP-18在胆汁对胃黏膜细胞损伤中的表达。方法健康雌、雄性各半SD大鼠44只,分成4组:全反流模型(DGR)组、结扎胆管(BDL)组、全反流模型+结扎胆管(DGR+BDL)组、对照(C)组。手术后12周处死大鼠。胃黏膜上皮细胞GES-1培养:以不同浓度(500μmol/L,1 000μmol/L)甘氨胆酸钠(GC),甘氨脱氧胆酸钠(GDC),甘氨鹅脱氧胆酸钠(GCDC),牛磺胆酸钠(TC),牛磺脱氧胆酸钠(TDC),牛磺鹅脱氧胆酸钠(TCDC),石胆酸(LCA),胆酸(CA)及脱氧胆酸(DCA),不同浓度(1 500μmol/L,2 000μmol/L)混合胆盐及混合胆酸作用后,用RT-PCR法检测大鼠胃黏膜、人胃黏膜上皮细胞AMP-18mRNA的表达。结果大鼠胃黏膜AMP-18表达,DGR较DGR+BDL、BDL及C组减少,差异有统计学意义(P<0.05),DGR+BDL与BDL组较对照组降低,但差异无统计学意义(P>0.05);各组GCDC、GDC、GC、TCDC、TDC、TC、DCA、LCA、CA、混合胆盐/酸作用于GES-1细胞后,GC实验组AMP-18表达虽随作用浓度的提高较对照组降低,但差异无统计学意义(P>0.05);余各组GES-1细胞AMP-18表达随作用浓度的提高均较对照组降低,差异有统计学意义(P<0.05)。结论 AMP-18在胆汁损伤的实验中,对胃肠黏膜损伤的修复有着重要作用。Objective To explore the effects of AMP-18 on gastric mucosa from rat model and cultured eternalized human gastric mucosa epithelium GES-1 cells.Methods Totally 44 healthy SD rats,half males and half females,were divided into 4 groups: duodenogastric reflux(DGR) group,bile duct ligation(BDL) group,DGR plus BDL group,and control group.Rats were killed 12 weeks after the operations.Cultured eternalized human gastric mucosa epithelium GES-1 cells were treated with media containing 6 different kinds of bile salts and 3 different kinds of bile acids and their mixture with different concentrations: glycochenodeoxycholate(GCDC),glycodeoxycholate(GDC),glycocholate(GC),taurochenodeoxycholate(TCDC),taurodeoxycholate(TDC),taurocholate(TC),lithocholic acid(LCA),cholic acid(CA),deoxycholic acid(DCA)(500 μmol/L,1 000 μmol/L),mixture of bile salts(DY),and mixture of bile acids(DS)(1 500 μmol/L,2 000 μmol/L).Rats in the control group were treated with normal media without bile salts or bile acids.RT-PCR was used to examine the expression of AMP-18 mRNA in rat gastric mucosa and human gastric mucosa epithelium.Results The expression of AMP-18 mRNA in the DGR group decreased significantly compared with that in the DGR plus BDL group,BDL group and control group(P0.05).The expression of AMP-18 mRNA in DGR plus BDL and BDL groups decreased compared with that in the control group,but there was no significant difference(P0.05).The expression of AMP-18 mRNA was lower in the GC group than in the control group,without significant difference(P0.05).The expression of AMP-18 mRNA in the other groups was significantly lower than that in the control group(P0.05) in a dose-dependent manner.Conclusion AMP-18 plays an important role in the restoration of gastric mucosal epithelial cell injury.
