摘  要:目的探讨调控因子Snail、Slug对E-钙黏蛋白的调控和参与人宫颈癌细胞转移侵袭的机制。方法对20例正常宫颈组织标本及85例宫颈鳞癌标本制成石蜡包埋切片,采用免疫组织化学染色方法检测Slug基因和间质标志物波形蛋白(Vimentin)的表达;采用免疫荧光化学染色检测转录因子Snail蛋白和上皮标志物E-cadherin的表达。结果在宫颈鳞癌组织中Slug蛋白和Vimentin的阳性表达率明显高于正常宫颈组织(P<0.05),随着临床分期增高,其阳性表达率有升高趋势,但未达到统计学差异(P>0.05)。随着组织分化级别下降和肿瘤淋巴结转移,其阳性表达率明显上升(P<0.05);Snail蛋白在鳞癌组织中荧光强度高于正常宫颈组织,浸润癌组织高于宫颈CIN组织(P<0.05),随着组织分化级别下降和肿瘤淋巴结转移,其荧光强度明显增强,但差异亦无统计学意义(P>0.05);E-cadherin在正常宫颈组织中荧光强度高于肿瘤组织,宫颈CIN组织高于浸润癌组织(P<0.05),随着组织分化级别下降和肿瘤淋巴结转移,其荧光强度明显减弱(P<0.05)。结论人宫颈组织上皮-间质转化在宫颈癌侵袭进展中可能发挥重要的作用,同时Snail、Slug信号通路可能参与了宫颈癌的侵袭转移过程。Objective To investigate the regulation of the transcription factors Snail and Slug on E-cadherin and their involvement in tumor epithelial-mesenchymal transition in human cervical cancer.Methods Totally 20 cases of normal cervical tissues and 85 cases of cervical cancer tissues were collected.The expression of Slug and Vimentin was detected by immunohistochemical S-P method and the expression of Snail and E-cadherin was detected by immunofluorescence method,respectively.Results The expression of Slug and Vimentin was significantly higher in cervical cancer than in normal cervical tissues(P0.05).Meanwhile,the positive expression rate of Slug and Vimentin increased with tumor clinical stage progression,but no significant correlation among different stages was observed(P0.05).With the tumor histological differentiation and lymph node metastasis,the positive expression rate of Slug and Vimentin also increased(P0.05).The expression of Snail was significantly higher in cervical cancer than in normal cervical tissues(P0.05).The positive expression of Snail in cervical tissues was significantly higher than in CIN tissues(P0.05).The expression of Snail was not significantly related to the tumor histological differentiation and lymph node metastasis(P0.05).With tumor clinical stage progression,the expression of E-cadherin was significantly lower in cervical cancer than in normal tissues(P0.05),the positive expression of E-cadherin in CIN was significantly higher than in the cancer(P0.05).With decline of tumor histological differentiation and lymph node metastasis,the expression of E-cadherin decreased(P0.05).Conclusion The epithelial-mesenchynal transition may play an important role in the progression of cervical carcinoma,and the transcription factors Snail and Slug with Vimentin may inhibit the cervical carcinoma through the related pathway.
